Molecular rationale for the impairment of the MexAB-OprM efflux pump by a single mutation in MexA

Pierpaolo Cacciotto et al.

Summary: Efflux pumps of the RND superfamily play a role in antibiotic resistance by expelling antibiotics from Gram-negative pathogens. This study investigated the stability of MexA, a membrane fusion protein, and the impact of a mutation on its function. The results show that MexA dimers are stable, but the mutation disrupts the protein's conformational equilibrium, making dimer or higher order assemblies unlikely.

In situ structure of the AcrAB-ToIC efflux pump at subnanometer resolution

Summary: This study presents the in situ structure of full-length Escherichia coli AcrAB-ToIC obtained by electron cryo-tomography, revealing the configuration and mechanism of this transmembrane efflux pump. The study suggests that AcrA acts as a bridge between the inner membrane and AcrB, enabling the transmission of drug-induced signals from AcrB to the ToIC channel within the pump.

Structural and Functional Diversity of Resistance-Nodulation-Cell Division Transporters

Philip A. Klenotic et al.

Summary: Multidrug resistant bacteria pose a global threat, making common infections increasingly difficult to eradicate. Research has shown that bacterial membrane proteins belonging to the resistance-nodulation-cell division superfamily play crucial roles in bacterial resistance to antibiotics. Studies have reviewed the assembly and function of bacterial inner membrane pumps, providing insights for the design and optimization of new efflux pump inhibitors to better treat difficult-to-cure infections.

Cryoelectron Microscopy Structures of AdeB Illuminate Mechanisms of Simultaneous Binding and Exporting of Substrates

Christopher E. Morgan et al.

Summary: Acinetobacter baumannii, a highly antibiotic-resistant Gram-negative pathogen, utilizes the AdeB multidrug efflux pump to resist a wide range of clinically relevant antimicrobial agents. The study of AdeB pump structures in the presence of ethidium bromide reveals that each AdeB protomer within the trimer can independently recognize and extrude drugs. This sheds light on the drug transport mechanism involving multiple multidrug-binding sites and various transient states of the AdeB membrane protein.

Structural and functional analysis of the promiscuous AcrB and AdeB efflux pumps suggests different drug binding mechanisms

Alina Ornik-Cha et al.

Summary: This study reveals the structure and drug-binding mechanism of the AdeB subunit of the Acinetobacter baumannii AdeABC efflux pump, as well as its differences with the Escherichia coli AcrB. The research shows that these efflux pumps can provide a deeper understanding of antibiotics and drug resistance.

Structure, Assembly, and Function of Tripartite Efflux and Type 1 Secretion Systems in Gram-Negative Bacteria

Ilyas Alav et al.

Summary: Tripartite efflux pumps and related type 1 secretion systems in Gram-negative organisms exhibit diverse functions, energization, and structural organization. This review highlights the role of periplasmic adaptor proteins (PAPs) in providing a common architectural scaffold and proposes a new integrated model of PAP-mediated communication within these systems.

Allosteric drug transport mechanism of multidrug transporter AcrB

Heng-Keat Tam et al.

Summary: Gram-negative bacteria maintain intrinsic resistance against noxious compounds through highly efficient efflux pumps. The AcrB drug efflux pump of E. coli, comprising three interdependent protomers, cycles through different states during cooperative catalysis. Structural analysis reveals novel drug-binding sites and transport pathways, suggesting allosteric modulation of drug transport in the presence of multiple drugs.

Ever-Adapting RND Efflux Pumps in Gram-Negative Multidrug-Resistant Pathogens: A Race against Time

Martijn Zwama et al.

Summary: The rise in multidrug resistance poses a major threat to human health globally due to the over-expression of multidrug efflux pumps and amino acid substitutions within the pumps themselves causing increased drug efflux efficiency. Studies on clinically, environmentally and laboratory-evolved Gram-negative bacterial strains have shown mutations in the RND-type pumps leading to decreased drug sensitivity. Understanding the evolution and variations in efflux pumps is crucial for the development of novel antibiotics and efflux pump inhibitors.
