The Celsr3-Kif2a axis directs neuronal migration in the postnatal brain

Janne Hakanen et al.

Summary: The planar cell polarity protein Celsr3 and the microtubule depolymerizing protein Kif2a cooperatively specify the directionality of neuroblasts tangential migration in the postnatal brain, providing new insights into the mechanisms of neuroblast migration.

KIF2A deficiency causes early-onset neurodegeneration

Nuria Ruiz-Reig et al.

Summary: KIF2A plays a critical role in the maturation, connectivity, and maintenance of neurons. Its lack disrupts microtubule dynamics, leading to aberrations in neuronal polarity, neuritogenesis, synaptogenesis, and axonal transport. Mutations in KIF2A result in abnormal electrophysiological characteristics, connectivity, and function in neurons.

Variants in KIF2A cause broad clinical presentation; the computational structural analysis of a novel variant in a patient with a cortical dysplasia, complex, with other brain malformations 3

Maiko Hatano et al.

Summary: CDCBM3 is a rare autosomal dominant syndrome caused by KIF2A gene mutation, characterized by posterior dominant agyria/pachygyria with severe motor dysfunction. A novel heterozygous mutation of NM_001098511.2:c.1298C>A [p.(Ser433Tyr)] was identified in a patient, leading to mild symptoms of CDCBM3 due to its location near the nucleotide binding domain pocket. Variants in KIF2A are associated with central nervous system and facial anomalies, suggesting a spectrum of KIF2A syndrome with variable severity.

Roles of developmentally regulated KIF2A alternative isoforms in cortical neuron migration and differentiation

Cansu Akkaya et al.

Summary: KIF2A is essential for dendritic arborization and radial migration of cortical neurons during brain development. Three Kif2a isoforms were studied, with only two supporting neuronal migration. A KIF2A mutation causing brain malformations led to changes in its interactome, including depletion of mitochondrial proteins.
