De novo design of a nanopore for single-molecule detection that incorporates a β-hairpin peptide

Summary: A de novo designed peptide with a beta-hairpin structure is able to assemble into beta-barrelled nanopores for single-molecule detection of polynucleotides and polypeptide chains. By manipulating amino acid sequences, artificial proteins and peptides with desired functionality can be developed. Redesigning the peptide to create monodisperse pores allows for detection of single polypeptide chains, showing potential for creating artificial nanopores adjustable to target molecules.

Development of Antimicrobial Stapled Peptides Based on Magainin 2 Sequence

Summary: Magainin 2, an antimicrobial peptide, relies on 17 amino acid residues for its antimicrobial activity, and derivatives with hydrocarbon stapling side chains can enhance this activity by stabilizing helical structures. The study found that hydrocarbon stapling significantly stabilizes helical structures of peptides, leading to improved antimicrobial activity without significant hemolytic effects. These findings suggest potential for developing novel antimicrobial agents based on Magainin 2 derivatives.

A Re-evaluation of the Free Energy Profiles for Cell-Penetrating Peptides Across DOPC Membranes

Summary: Cell penetrating peptides (CPPs) have the potential to transport therapeutic agents to their targets with low cytotoxicity and high efficiency. However, the complexity of cellular uptake mechanisms and experimental variations hinder quantitative analysis and comparison of data. This study used molecular dynamics simulations to investigate the translocation of seventeen CPPs, revealing that positively charged residues cause repulsion and hydrophobic residues enhance bilayer interaction.
