Interleukin-29 Accelerates Vascular Calcification via JAK2/STAT3/BMP2 Signaling

Summary: Vascular calcification (VC), characterized by the osteogenic transdifferentiation of vascular smooth muscle cells, is associated with enhanced cardiovascular morbidity and mortality. Inflammation plays a key role in VC initiation and progression. IL-29, a member of type III interferon, is implicated in the pathogenesis of autoimmune diseases. This study evaluated the role of IL-29 in VC and its underlying inflammatory mechanisms.

Inhibition of alkaline phosphatase impairs dyslipidemia and protects mice from atherosclerosis

Summary: This study found that the activation of tissue-nonspecific alkaline phosphatase (TNAP) precedes and predicts calcification in atherosclerotic plaques, and TNAP inhibition prevents plaque calcification and protects arteries from atherosclerosis. The study also discovered that TNAP inhibition reduces blood levels of cholesterol and triglycerides, and ameliorates plaque calcification, thereby protecting arteries.

Summary: This study investigated the impact of Klotho deficiency on arterial structure and found that it leads to severe arterial calcification and elastin fragmentation. Klotho deficiency upregulates the expression of BMP2 and RUNX2, and is independent of hyperphosphatemia in causing arterial calcification. Additionally, Klotho deficiency induces osteoblastic transition in SMCs and activates the BMP2-RUNX2 signaling pathway.
