Metabolic Perturbation and Synthetic Biology Strategies for Plant Terpenoid Production-An Updated Overview

Summary: Terpenoids, with vast structural diversity, are high-value specialized metabolites widely used in cosmetics, foods, and pharmaceuticals. Recent advancements in heterologous production platforms and metabolic engineering technologies have overcome challenges related to the bioavailability of terpenoids in natural sources. Studies have shown that subcellular localization of precursor pool and introduced enzymes are crucial for increasing targeted terpenoid production in plants.

Metabolic engineering of tomato fruit enriched in L-DOPA

Summary: The study successfully engineered tomato fruit enriched in L-DOPA through overexpression of BvCYP76AD6, and further increased L-DOPA levels by elevating the expression of the metabolic master regulator MYB12. The research also revealed new roles for L-DOPA in plants, impacting fruit quality parameters and providing opportunities for the development of new biological sources of L-DOPA.

Engineering Betalain Biosynthesis in Tomato for High Level Betanin Production in Fruits

Summary: Betalains are pigments found in plants of the Caryophyllales order, and betanin is a red pigment made from tyrosine by a biosynthetic pathway consisting of cytochrome P450, L-DOPA dioxygenase, and glucosyltransferase. Recent studies have shown that the high amount of betanin in red beets is due to a duplicated plastidic arogenate dehydrogenase gene with relaxed feedback inhibition by tyrosine. By reconstituting the betanin biosynthetic pathway in tomato plants with or without the BvADHα gene, researchers have demonstrated that the gene can increase betanin production. Transformed tomatoes produced only betacyanins, giving the tomato juice a bright purple-fuschia color.
