QTL mapping reveals candidate genes for main agronomic traits in Luffa based on a high-resolution genetic map

Genome-Wide Association Studies Reveal Candidate Genes Related to Stem Diameter in Cucumber (Cucumis sativus L.)

Summary: This study measured the stem diameter of cucumber core germplasms and conducted a genome-wide association study. It identified eight loci significantly associated with cucumber stem diameter, and five of them were considered reliable as they were repeatedly detected in multiple seasons. Further analysis revealed five genes that showed variations in amino acid sequence between thick stem lines and thin stem lines, and the expression pattern of these genes also differed significantly. This study provides a foundation for gene cloning and molecular mechanism study of cucumber stem development.

Development of KASP Markers and Identification of a QTL Underlying Powdery Mildew Resistance in Melon (Cucumis melo L.) by Bulked Segregant Analysis and RNA-Seq

Summary: A novel QTL named qCmPMR-12 for powdery mildew resistance in melon was identified on chromosome 12, ranging from 22.0 Mb to 22.9 Mb. The candidate gene MELO3C002434 encoding an ankyrin repeat-containing protein was suggested to be associated with resistance to PM. Furthermore, 15 polymorphic SNPs were successfully converted to KASP markers with high significance (P < 0.0001).

It was found that CsTFL1d can partially rescue determinate growth of CsTFL1(del) through induction by long-day and high-temperature signals. Biochemical analysis showed that CsTFL1d interacts with CsNOT2a and functions in a similar regulatory mechanism as CsTFL1.

A mutation in LacDWARF1 results in a GA-deficient dwarf phenotype in sponge gourd (Luffa acutangula)

Summary: The dwarfism gene LacDWARF1 was mapped to a specific region on chromosome 05 using BSA-Seq and comparative genomics analyses. The study identified an insertion in the gibberellin metabolism-related gene Lac05g019500 in the dwarf mutant WJ209, leading to transcription errors. The dwarf phenotype of WJ209 was found to be due to a deficiency in bioactive GAs, which could be restored by exogenous GA(3) treatment.
