Synergistic enzyme cocktail between levansucrase and inulosucrase for superb levan-type fructooligosaccharide synthesis

Summary: In this study, a new strategy was proposed to improve the production of levan-type fructooligosaccharides (L-FOS) by using a mixed reaction of inulosucrase (ISC) and levansucrase (LSC). The presence of ISC in the LSC reaction significantly increased the production of L-FOS as the main products. The ratio between ISC and LSC also affected the distribution of L-FOS, with more ISC leading to the synthesis of short-chain L-FOS and more LSC promoting the synthesis of longer-chain L-FOS. The addition of trisaccharide mixtures obtained from ISC or LSC reactions enhanced L-FOS synthesis in the LSC reaction. Analysis of these trisaccharides revealed that most oligosaccharide species were similar, with 1-kestose being the major one. The results were further supported by molecular dynamics simulations.

Summary: The novel alpha-glucosidase from Weissella cibaria BBK-1 has been biochemically and structurally characterized in this study, showing specific hydrolytic activity on short-chain oligosaccharides and strong transglycosylation activity in the presence of maltose. Size-exclusion chromatography and X-ray crystal structures revealed that WcAG forms a homodimer with a unique structure, leading to its substrate preference.
