Summary: Loss of genome stability can lead to reduced fitness, fertility, and a high mutation rate. Through a genetic screen in Arabidopsis thaliana, it was found that zebularine induces DNA damage by interfering with the maintenance of CG DNA methylation, likely through enzymatic DNA-protein crosslinks of MET1 and zebularine-containing DNA. Zebularine-induced DNA-protein crosslinks accumulate preferentially in specific regions in chromocenters, offering a new avenue for studying genome stability and DPC repair in plants.
