JASPAR 2022: the 9th release of the open-access database of transcription factor binding profiles

Jaime A. Castro-Mondragon et al.

Summary: JASPAR is an open-access database containing manually curated, non-redundant transcription factor binding profiles across six taxonomic groups. The latest release introduces new profiles for plants, vertebrates, urochordates, and insects, as well as new tools for TFBS enrichment analysis and data access. The database can be accessed through various platforms, including the JASPAR website, RESTful API, R/Bioconductor data package, and a new Python package pyJASPAR.

Tissue-specific analysis of Fgf18 gene function in palate development

Minghui Yue et al.

Summary: The study demonstrates that Fgf18 expression in neural crest-derived craniofacial mesenchyme plays a critical role in the development of the mandible and multiple craniofacial bones, while Fgf18 expression in the palatal mesenchyme is dispensable for palatogenesis.

Creb5 establishes the competence for Prg4 expression in articular cartilage

Cheng-Hai Zhang et al.

Summary: Zhang et al. identify Creb5 as a critical transcription factor required for the induction of Prg4 expression by TGF beta and EGFR signaling. They demonstrate that Creb5 directly binds to two proximal regulatory elements of the Prg4 promoter, working in conjunction with a distal regulatory element to drive Prg4 induction by TGF beta. These findings offer new insights into the regulatory mechanisms underlying Prg4 expression in superficial zone articular chondrocytes.

CRISPRi-mediated functional analysis of NKX2-1-binding sites in the lung

William D. Stuart et al.

Summary: NKX2-1, a transcription factor, plays a crucial role in lung pathophysiology, but its precise functional significance in non-proximal regions remains unclear. Stuart and Fink-Baldauf's study using CRISPRi method examines 19 NKX2-1 binding sites in genes relevant to breathing, immune defense, and tumorigenesis, revealing differential importance in gene expression regulation.
