Multiomics reveal the central role of pentose phosphate pathway in resident thymic macrophages to cope with efferocytosis-associated stress

Summary: This study characterizes the metabolic features of tissue-resident macrophages and reveals the preferential activation of the pentose phosphate pathway (PPP) in thymic macrophages. The PPP is essential for the efferocytosis of dying thymocytes and blocking it leads to decreased efferocytosis, which can be rescued by reactive oxygen species (ROS) scavengers.

Summary: Macrophages in the testis and epididymis originate from different stages of cell development and can self-maintain independently of bone marrow hematopoietic cells in a steady state. However, during radiation-induced macrophage ablation or infection, bone marrow-derived cells are recruited to the testis and epididymis, leading to an inflammatory response.

Summary: This review focuses on the fibrogenic roles of increased TGF-beta activities and underlying signaling mechanisms in chronic diseases marked by fibrosis. Insight into these roles and mechanisms of TGF-beta as a universal driver of fibrosis has stimulated the development of therapeutic interventions to attenuate fibrosis progression, based on interference with TGF-beta signaling. The promise of these interventions in preclinical and clinical settings will be discussed.
