Exercise triggers CAPN1-mediated AIF truncation, inducing myocyte cell death in arrhythmogenic cardiomyopathy

Stephen P. Chelko et al.

Summary: Research has shown that in a mouse model mimicking arrhythmogenic cardiomyopathy, myocardial cell death and heart dysfunction are primarily affected by calcium overload and oxidation of apoptosis-inducing factor (AIF). Preventing CAPN1-induced AIF truncation or inhibiting the binding of AIF to the nuclear chaperone PPIA may prevent myocardial cell death and slow down disease progression in ACM and other cardiomyopathies.

Optic nerve regeneration screen identifies multiple genes restricting adult neural repair

Jane A. Lindborg et al.

Summary: Adult mammalian central nervous system (CNS) trauma leads to persistent neurological deficits due to minimal axonal regeneration. In this study, suppression of 40 genes was found to promote retinal ganglion cell axon regeneration after optic nerve crush. Loss of interleukin 22 (IL-22) contributes to early inflammatory responses, activation of Stat3, DLK pathways, and upregulation of neuron-intrinsic regeneration-associated genes.

Myocardial Infarction Induces Cardiac Fibroblast Transformation within Injured and Noninjured Regions of the Mouse Heart

Haisam Shah et al.

Summary: Heart failure is associated with pathological remodeling of the myocardium, with activated cardiac fibroblasts playing a crucial role in fibrosis and scar formation. Mass spectrometry analysis revealed global activation of cardiac fibroblasts post-injury, with differential protein expression between fibroblasts from remote and infarct regions of injured hearts. These findings suggest that myocardial infarction induces significant changes to the cardiac fibroblast proteome, with differences reflecting proximity to the site of injury.

Mitochondrial health is enhanced in rats with higher vs. lower intrinsic exercise capacity and extended lifespan
