Microcrystalline silica particles induce inflammatory response via pyroptosis in primary human respiratory epithelial cells

Summary: This study reveals that DNA methylation and overexpression of MBD2 are associated with fibroblast differentiation and pulmonary fibrosis in idiopathic pulmonary fibrosis (IPF) and a mouse model. TGF-β1 induces global DNA hypermethylation and Mbd2 overexpression in a TβRI/Smad3 dependent manner, leading to repression of Erdr1 expression and promotion of fibroblast differentiation into myofibroblasts. Enhancing Erdr1 expression can significantly reverse pulmonary fibrosis.

Crosstalk between epitranscriptomic and epigenetic mechanisms in gene regulation

Summary: Epigenetic modifications play a crucial role in gene expression regulation through modifications on genomic DNA, histones, and more recently, mRNA. These modifications, especially the common N6-methyladenosine (m6A) methylation, not only influence RNA stability and translation efficiency, but also interact with histone/DNA epigenetic mechanisms to specify transcriptional outputs and recruit chromatin modifiers. This review focuses on dissecting the feedback circuits orchestrated by m6A, which regulate histone modifications and the activity of regulatory RNAs, such as long noncoding RNA and chromosome-associated regulatory RNA. The evidence suggests that m6A acts as a versatile checkpoint linking different layers of gene regulation.

LncRNA UCA1 regulates silicosis-related lung epithelial cell-to-mesenchymal transition through competitive adsorption of miR-204-5p

Summary: This study observed the changes and effects of UCA1 and miR-204-5p in silica-induced mouse silicosis model and in vitro EMT model of A549 cells. It was found that both models showed the presence of EMT process, and UCA1 was upregulated in the in vitro model. Silencing UCA1 inhibited EMT process, while intervention of miR-204-5p regulated EMT.

Macrophage derived miR-7219-3p-containing exosomes mediate fibroblast trans-differentiation by targeting SPRY1 in silicosis

Summary: Silicosis is a serious occupational disease characterized by inflammation, fibroblast activation, and extracellular matrix deposition in the lung. Macrophage-derived exosomes have been found to play a role in the development of silicosis by transferring microRNAs (miRNAs). In this study, the researchers identified the up-regulation of miR-7219-3p in both macrophages and their exosomes after silica exposure, as well as in a silicotic mice model. They found that miR-7219-3p promoted fibroblast to myofibroblast trans-differentiation, cell proliferation, and migration. The target gene of miR-7219-3p, Spouty1 (SPRY1), was confirmed to modulate the Ras/ERK/MAPK signaling pathway, and manipulating SPRY1 expression affected fibroblast to myofibroblast trans-differentiation via this pathway. Inhibition of exosomal miR-7219-3p partially suppressed fibroblast to myofibroblast trans-differentiation and silica-induced pulmonary fibrosis. These findings suggest that exosomal miR-7219-3p could be a potential therapeutic target for silicosis.

Long non-coding RNA lnc-SNHG1 was found to be highly expressed in fibrotic lung tissues and TGF-beta 1-treated fibroblasts, promoting fibroblast migration, invasion, and fibrotic molecule secretion. It was discovered that lnc-SNHG1 may regulate pulmonary fibrosis through the miR-326/SP1 axis.

Anti-fibrotic effects and the mechanism of action of miR-29c in silicosis
