Bioengineered Living Bone Grafts-A Concise Review on Bioreactors and Production Techniques In Vitro

Paulina Kazimierczak et al.

Summary: Bone fractures carry significant risks, and proper bone healing methods are crucial for successful recovery. Bone tissue engineering (BTE) offers promising approaches to restore bone defects by combining bone scaffolds with cells and healing promotive factors. This review highlights the latest findings in biomaterials, cells, and techniques used for producing living bone grafts in vitro, with particular attention to bioreactor systems such as perfusion, rotating, and spinner flask bioreactors. Despite limitations, bioreactor systems prove to be excellent platforms for bioengineering living bone grafts for bone fracture regeneration.

Comparison of Growth and Chemical Profile of Diatom Skeletonema grevillei in Bioreactor and Incubation-Shaking Cabinet in Two Growth Phases

Roberta Frleta et al.

Summary: Marine diatoms, such as Skeletonema grevillei, are a valuable source of compounds with various applications in industries such as food, cosmetics, pharmaceuticals, and biodiesel production. This study compared the growth, chemical profiles, and antioxidant activity of S. grevillei cultivated in a bioreactor and incubation-shaking cabinet. Results showed that the bioreactor system promoted faster growth and higher extract yield compared to the incubation-shaking cabinet. Oleamide was identified as the dominant compound, and the S. grevillei extract from the bioreactor after 312 h exhibited the highest antioxidant activity.

Nano-Structured Ridged Micro-Filaments (≥100 μm Diameter) Produced Using a Single Step Strategy for Improved Bone Cell Adhesion and Proliferation in Textile Scaffolds

Nemeshwaree Behary et al.

Summary: Textile scaffolds made through textile processing lack nano-topographical features to improve bone cell adhesion and proliferation. In this study, biodegradable nanostructured PLLA microfilaments with diameters of 100μm and 230μm were fabricated using a single step melt-spinning process, and showed excellent biological performance better than smooth fibers.

The Mineralization of Various 3D-Printed PCL Composites

Artem Egorov et al.

Summary: In this study, different calcification methods for collagen and collagen coatings were compared for their suitability for 3D printing and production of collagen-coated scaffolds. HA directly in the collagen solution resulted in increased viscosity and led to clumping on the scaffolds. The incubation and ALP coating methods resulted in increased HA layer thickness, while no coating on the collagen layer was observed with the poly-L-aspartic acid method.

Effect of cyclic mechanical loading on immunoinflammatory microenvironment in biofabricating hydroxyapatite scaffold for bone regeneration

Penghui Zhang et al.

Summary: The study demonstrated that mechanical loading can influence the inflammatory responses and osteogenic differentiation of MSCs. Biofabrication of scaffolds with mechanical loading led to downregulation of inflammatory mediators and upregulation of ALP and COL1A1. The ECM-based scaffolds biofabricated with mechanical loading before freeze-drying facilitated M2 polarization of macrophages and bone regeneration, showing promise as a strategy for bone biomaterials.
