IMPDH1 retinal variants control filament architecture to tune allosteric regulation

Anika L. Burrell et al.

Summary: A series of cryo-EM structures of human IMPDH1 variants reveal polymorphic filaments. Blindness-associated mutations in IMPDH1 are characterized and half disrupt feedback inhibition. These findings provide a foundation for understanding the role of IMPDH1 in retinal function and disease.

Structural basis for the catalytic activity of filamentous human serine beta-lactamase-like protein LACTB

Minghui Zhang et al.

Summary: In this study, the cryoelectron microscopy structures of the human LACTB (hLACTB) were determined. It was found that the assembly of hLACTB filaments requires three interfaces and the formation of higher order helical structures facilitates its cleavage activity. Furthermore, the study showed that the middle region is necessary for substrate hydrolysis but not for filament formation. The inhibitor-bound structure suggests that hLACTB may cleave peptide bonds adjacent to aspartic acid residues.

Structure and functionality of a multimeric human COQ7:COQ9 complex

Mateusz Manicki et al.

Summary: This study presents structure-function analyses of a complex comprising COQ7 and COQ9, revealing their roles in the synthesis of CoQ. The findings suggest that COQ7 and COQ9 interact to form a complex that facilitates the translocation of CoQ intermediates from the membrane to the protein's lipid-binding sites.

Structural basis for isoform-specific inhibition of human CTPS1

Eric M. Lynch et al.

Summary: CTPS1 is essential for an effective immune response, with deficiency leading to severe immunodeficiency. CTPS1 expression is up-regulated in activated lymphocytes to meet the increased demand for nucleic acid and lipid synthesis. Selective inhibition of CTPS1 could be beneficial for treating immune disorders and lymphocyte cancers, with recent small-molecule inhibitors showing promise in inhibiting proliferation of human T cells.

Membrane binding controls ordered selfassembly of animal septins

Agata Szuba et al.

Summary: Septins interact with flat lipid membranes through electrostatic interactions and form mechanically rigid filamentous meshworks, which require membrane binding for their ordered assembly. The stacking of filaments is mediated by C-terminal coiled coils on specific subunits.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

Mitochondrial compartmentalization: emerging themes in structure and function

Joseph C. Iovine et al.

Summary: Compartmentalization within cellular structures refers to the spatial segregation of macromolecules, metabolites, and biochemical pathways, bridging organellar structure and function. Mitochondria, with their morphological complexity and dynamic polymorphism, undergo morphogenesis events that must be considered both spatially and temporally. Disturbance in mitochondrial compartmentalization can lead to dysfunction associated with heritable and aging-related diseases.
