Symmetry-breaking of animal cytokinesis

Kenji Sugioka

Summary: Cytokinesis is a mechanism that separates dividing cells using a supramolecular structure called the contractile ring. The symmetry-breaking modes of cytokinesis, including unilateral cytokinesis, asymmetric cell division, and oriented cell division, play important roles in tissue patterning and morphogenesis. While the classic mechanism involves the geometric relationship between the mitotic spindle and cell cortex, emerging evidence suggests that a concerted flow of compressional cell-cortex materials can also drive spatial cytokinesis control.

Cell size and polarization determine cytokinesis furrow ingression dynamics in mouse embryos

Lia Mara Gomes Paim et al.

Summary: This study investigates the dynamics of furrow ingression in early mouse embryos during cell division. It finds that cell size is an important determinant of furrow ingression speed, and the emergence of cell polarity and assembly of the apical domain negatively regulate furrow ingression on one side. The study reveals that cell polarity directly regulates the recruitment of cytokinetic machinery in a cell-autonomous manner.

Cytokinetic diversity in mammalian cells is revealed by the characterization of endogenous anillin, Ect2 and RhoA

Summary: Cytokinesis is a crucial process that requires the assembly and ingression of an actomyosin ring to physically separate daughter cells. By studying the localization of key molecules during cytokinesis in live human cells, differences in cytokinetic diversity were observed among different cell types.

Plastin and spectrin cooperate to stabilize the actomyosin cortex during cytokinesis

Ana Filipa Sobral et al.

Summary: The study demonstrates a synergy between the small rigid crosslinker plastin and the large flexible crosslinker spectrin in the C. elegans embryo, showing that their combined action stabilizes the actomyosin network in cytokinesis. Co-inhibition of plastin and beta H-spectrin results in cytokinesis failure, highlighting the importance of crosslinker interplay for network integrity.

Summary: During cell division, the GEF ECT2 activates RhoA in a narrow zone at the cell equator. The BRCT domains, especially BRCT1 and BRCT2, play crucial roles in controlling ECT2 activity spatially and temporally to ensure successful cell division.

Inhibition of polar actin assembly by astral microtubules is required for cytokinesis

Anan Chen et al.

Summary: During cell division, the opposing regulation of beta-actin and gamma-actin networks is crucial for successful cytokinesis. Depletion of gamma-actin at cell poles through localized deactivation of DIAPH1, and production of a beta-actin contractile ring at the cell equator, are necessary for the completion of cell division.

Quantitative live-cell imaging and computational modeling shed new light on endogenous WNT/CTNNB1 signaling dynamics

Saskia M. A. de Man et al.

Summary: WNT/CTNNB1 signaling regulates tissue development and homeostasis in multicellular animals by affecting the dynamic distribution of CTNNB1 through regulatory nodes including the destruction complex, nucleocytoplasmic shuttling, and nuclear retention, as revealed by an integrated experimental and computational approach. The study found that a substantial fraction of CTNNB1 resides in slow-diffusing cytoplasmic complexes, which undergo a major reduction in size when the pathway is hyperactivated. The research provides quantitative insight into endogenous protein behavior and sheds light on the molecular mechanism underlying WNT/CTNNB1 signaling.

CRISPR-Cas tools to study gene function in cytokinesis

Mathieu C. Husser et al.

Summary: Cytokinesis is the process of separating a cell into two daughter cells at the end of mitosis, with most knowledge coming from overexpression studies. Gene editing can introduce functional mutations or fluorescent probes directly into a gene locus to circumvent issues with protein function interpretation, but its use in the field of cytokinesis is just beginning.
