Neuroinflammation in perioperative neurocognitive disorders: From bench to the bedside

Yang Liu et al.

Summary: Perioperative neurocognitive disorders (PNDs) are common complications in elderly patients, characterized by cognitive decline after anesthesia and surgery. Neuroinflammation, particularly in the elderly, has been identified as a major cause, with the activation of glial cells playing a significant role in the inflammatory responses in the central nervous system (CNS). Clinical and preclinical studies have provided evidence from various levels to understand PNDs and their potential therapeutic targets, but further research is needed to investigate the signaling pathways and their relationship with future neurocognitive dysfunction.

Elamipretide (SS-31) Improves Functional Connectivity in Hippocampus and Other Related Regions Following Prolonged Neuroinflammation Induced by Lipopolysaccharide in Aged Rats

Yang Liu et al.

Summary: Our study found that inhibiting hippocampal neuroinflammation not only reduces inflammatory responses in the hippocampus but also improves brain functional connectivity in regions related to the hippocampus. Early anti-inflammatory treatment with SS-31 has a long-lasting effect on reducing the impact of LPS-induced neuroinflammation.

Association Between Incident Delirium Treatment With Haloperidol and Mortality in Critically Ill Adults*

Matthew S. Duprey et al.

Summary: The study suggests that using haloperidol to treat incident delirium and its symptoms in the ICU may be associated with a dose-dependent improvement in survival. However, the protective effect of treatment haloperidol decreases when administered later in the ICU course. Future randomized trials are needed to confirm these findings.

Small-World Networks and Their Relationship With Hippocampal Glutamine/Glutamate Concentration in Healthy Adults With Varying Genetic Risk for Alzheimer's Disease

Hui Zhang et al.

Summary: The ApoE4 allele is the most common gene polymorphism related to Alzheimer's disease, and it remains unclear whether ApoE4 status affects hippocampal neuromodulation and brain network topology. This study found that healthy ApoE4 carriers exhibit poorer local interconnectivity and a close relationship between glutamate and small-world network properties in these carriers may reflect a compensatory response to impaired network efficiency.

State-independent and state-dependent patterns in the rat default mode network

Wei Jing et al.

Summary: Resting-state studies have traditionally assumed constant functional connectivity between brain regions, but recent evidence suggests dynamic fluctuations in higher-order regions such as the default mode network. The neural activities underlying this dynamic connectivity are not well understood, but factor analysis can reveal hidden patterns within the brain. Different spatial functional connectivity patterns have been identified based on temporal dynamics, some state-dependent and others state-independent. These spatial patterns with dynamic temporal contributions offer a flexible framework for integrating information to support cognition and behavior.

Summary: Recent studies on the mouse PFC have utilized the experimental toolbox available for this species, revealing cellular and structural differences in the PFC and neuronal activity related to theories about PFC functions. These data-rich studies in mice are expected to provide insights into the general prefrontal architecture and mechanisms underlying cognitive aspects of organized actions.

Dementia with Lewy bodies: association of Alzheimer pathology with functional connectivity networks

Julia Schumacher et al.

Summary: The presence of concurrent Alzheimer's disease pathology in patients with DLB is associated with more severe loss of default mode network connectivity. Additionally, higher functional connectivity between brain regions is related to higher tau pathology levels in cognitively normal, Alzheimer's disease, and DLB patients.

The default mode network in cognition: a topographical perspective

Jonathan Smallwood et al.

Summary: The default mode network (DMN) consists of widely distributed brain regions in the parietal, temporal, and frontal cortex, showing reduced activity during attention-demanding tasks and increased activity during complex cognition tasks. DMN is located in regions furthest away from sensory and motor systems in the cortex. Leveraging knowledge of the topographic characteristics of DMN can enhance understanding of its contribution to cognition and behavior.
