The tumour microenvironment shapes innate lymphoid cells in patients with hepatocellular carcinoma

Bernd Heinrich et al.

Summary: Our study shows that the cytokine milieu in liver cancer controls the composition of ILCs and the outcome of HCC. Specific cytokine changes can alter ILC composition in the tumor, induce plasticity, and modify ILC function.

Cytotoxic granzyme C-expressing ILC1s contribute to antitumor immunity and neonatal autoimmunity

Briana G. Nixon et al.

Summary: Innate lymphocytes are important components of the cellular immune system. They can defend the host against various challenges and can lead to immunopathology when dysregulated. This study found that granzyme C, a cytolytic molecule, was expressed in a specific type of innate lymphoid cells (ILC1s) in the liver and salivary gland of mice. These granzyme C-expressing ILC1s were derived from ILC progenitors and did not convert into NK cells, ILC2s, or ILC3s. Granzyme C marked a maturation state of ILC1s, and its expression in ILC1s required specific transcription factors and support from transforming growth factor-beta signaling. Depleting ILC1s in a mouse breast cancer model resulted in accelerated tumor growth. IL-15 stimulation induced granzyme C expression in ILC1s, enabling them to exhibit perforin-mediated cytotoxicity. However, constitutive activation of STAT5 in granzyme C-expressing ILC1s led to lethal perforin-dependent autoimmunity in neonatal mice. This study broadens the understanding of the function of ILC1s beyond helper-like lymphocytes.

Group 1 innate lymphoid-cell-derived interferon-γ maintains anti-viral vigilance in the mucosal epithelium

John P. Shannon et al.

Summary: The oropharyngeal mucosa serves as a primary site for pathogen entry and viral replication. During acute oral mucosal viral infection, ILC1s play a crucial role in providing early protection through IFN-γ production, enhancing resistance against oral mucosal viruses.

Human primed ILCPs support endothelial activation through NF-κB signaling

Giulia Vanoni et al.

Summary: ILCs play key roles in early immune responses, but their involvement in cancer is still poorly understood. ILCPs were found to induce upregulation of adhesion molecules in endothelial cells, promoting immune cell adhesion and potentially inhibiting tumor growth.

CD127+CD94+innate lymphoid cells expressing granulysin and perforin are expanded in patients with Crohn's disease

L. Krabbendam et al.

Summary: The study identifies two populations of CD127+CD94+ ILCs in the intestinal lamina propria, designated as population A and B, with population B expressing granulysin. Granulysin-expressing effector CD127+CD94+ ILCs are shown to be expanded in patients with Crohn's disease. The overlapping phenotypic markers between ILC1 and NK populations have impacted the specific analysis of these immune cell populations.

Conventional NK cells and tissue-resident ILC1s join forces to control liver metastasis

Laura Ducimetiere et al.

Summary: This study found that innate lymphocyte subpopulations cNKs and trILC1s play nonredundant roles in controlling liver metastasis seeding and outgrowth, respectively, and may contribute to developing therapies involving hepatic innate cells.
