Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

A Progressive Somatic Cell Niche Regulates Germline Cyst Differentiation in the Drosophila Ovary

Jingyuan Shi et al.

Summary: In the germarium of the Drosophila ovary, a population of somatic escort cells can be divided into two successive domains, with the anterior escort cells producing ecdysone to promote cell division and the posterior escort cells responding to ecdysone signaling to regulate cell adhesion. This distribution of escort cells is independent of the germline but dependent on JAK/STAT signaling activity, orchestrating cystoblast division and differentiation towards egg chamber formation.

Polyploid mitosis and depolyploidization promote chromosomal instability and tumor progression in a Notch-induced tumor model

Xian-Feng Wang et al.

Summary: This study shows that upregulation of the Notch signaling pathway induces polyploid mitosis in tumor cells in a Drosophila solid-tumor model, and DNA-damage response genes are upregulated in these tumors and required for ploidy-reduction division. These findings suggest that polyploidy and associated cell-cycle variants play a critical role in increasing tumor-cell heterogeneity and genome instability during cancer progression.

A single-cell atlas reveals unanticipated cell type complexity in Drosophila ovaries

Maija Slaidina et al.

Summary: Using single-cell RNA sequencing, researchers characterized all cell types of the stem cell compartment and early follicles of the Drosophila ovary, identifying specific markers for germ cell differentiation states and somatic cell types. They discovered a diverse population of escort cells with unique molecular expressions in discrete anatomical positions, suggesting complex micro-environments that support germ cell differentiation. This study provides a comprehensive resource for gene expression, cell-specific markers, spatial coordinates, and functional predictions for 34 ovarian cell types and subtypes.

Modeling Notch-Induced Tumor Cell Survival in the Drosophila Ovary Identifies Cellular and Transcriptional Response to Nuclear NICD Accumulation

Allison Jevitt et al.

Summary: Research has shown that the Notch signaling pathway is dysregulated in many cancer types, often through nuclear accumulation of the active receptor NICD. Using a tumor model in Drosophila ovarian follicular epithelium, it was found that NICD accumulation contributes to tumor growth and reduced apoptosis. Key genes involved in the response to NICD accumulation during tumorigenesis were identified through bulk RNA sequencing.
