A p53-phosphoinositide signalosome regulates nuclear AKT activation

Mo Chen et al.

Summary: The study reveals a novel mechanism of interaction between nuclear p53 and the PI3K-AKT pathway, which is distinct from the canonical membrane-localized pathway. It shows that genotoxic stress activates nuclear AKT through a p53-dependent mechanism, leading to the inhibition of DNA damage-induced apoptosis. This finding highlights the therapeutic relevance of the nuclear p53-phosphoinositide signalosome, as it is insensitive to currently available PI3K inhibitors.

TSC1 binding to lysosomal PIPs is required for TSC complex translocation and mTORC1 regulation

Katharina Fitzian et al.

Summary: The TSC complex serves as a critical negative regulator of small GTPase Rheb and mTORC1 in cellular stress signaling. TSC1 has been characterized structurally and functionally, showing its vital role in the function of the TSC complex and regulation of mTORC1 signaling.

Akt Isoforms: A Family Affair in Breast Cancer

Alakananda Basu et al.

Summary: This article discusses the importance of breast cancer in women in the United States and the role of Akt isoforms in the development of breast cancer. While Akt isoforms share similar structures, they exhibit redundant, distinct, and opposite functions in breast cancer. Understanding the isoform-specific function of Akt is critical for effectively targeting the Akt signaling pathway in cancer therapy.
