Defining the regenerative effects of native spider silk fibers on primary Schwann cells, sensory neurons, and nerve-associated fibroblasts

Flavia Millesi et al.

Summary: The study highlights the potential of native spider silk in promoting nerve regeneration, with insights needed on the in vivo regenerative properties of recombinant silk fibers. Results demonstrate that native spider silk supports attachment, proliferation, and characteristic behaviors of various cell types; axonal out-growth of DRG neurons occurs along SCs and SCs migrate along the silk fibers, achieving reported distance for regenerating axons.

Spider silk nerve graft promotes axonal regeneration on long distance nerve defect in a sheep model

T. Kornfeld et al.

Summary: This study investigated the effects of spider silk materials on axonal regeneration in peripheral nerve defects. The results showed that the spider silk nerve implant supported axonal regeneration comparable to autologous nerve grafts, indicating that spider silk nerve conduits could be an important approach for long nerve defects.

Summary: The multiomics approach revealed the essential roles of the MaSp3B spidroin and SpiCE proteins in achieving the mechanical properties of dragline silk of golden orb-weaver spiders. The combination of these components, along with MaSp1 and MaSp2, is crucial for realizing the unmatched toughness and strength of the silk.

Engineering Silk Fibroin-Based Nerve Conduit with Neurotrophic Factors for Proximal Protection after Peripheral Nerve Injury

Cristiana R. Carvalho et al.

Summary: Artificial nerve conduits made of silk fibroin are developed to incorporate neurotrophic factors for effective nerve regeneration. The controlled delivery of nerve growth factor and GDNF in these conduits is demonstrated to promote retrograde neuroprotection and motor nerve reinnervation, showing significant improvement in nerve regeneration after injury.
