An autoimmune stem-like CD8 T cell population drives type 1 diabetes

Summary: CD8 T cell-mediated autoimmune diseases are caused by the breakdown of self-tolerance mechanisms. This study identified a stem-like autoimmune progenitor population in the pancreatic draining lymph node, which continuously produces autoimmune mediators that migrate to the pancreas and destroy beta-cells. Strategies targeting this autoimmune progenitor pool could emerge as novel immunotherapeutic interventions for type 1 diabetes.

Summary: Multiple sclerosis (MS) is an autoimmune disease where autoreactive lymphocytes attack the central nervous system. Epstein-Barr virus (EBV) infection has been linked to MS, with molecular mimicry between EBV nuclear antigen 1 (EBNA1) and central nervous system protein GlialCAM. EBNA1 immunization exacerbates disease in MS mouse models and anti-EBNA1 and anti-GlialCAM antibodies are prevalent in MS patients, providing a mechanistic link between MS and EBV and guiding the development of new MS therapies.

Additionally, Round-shaped CCMVTT-VLPs led to prominent formation of splenic germinal centers and polarization of T cell response towards Th1. This is the first study to compare the draining kinetics and immunogenicity of VLPs forming nano-sized icosahedra or micrometer-sized rods.

Lymph Node-Targeted Synthetically Glycosylated Antigen Leads to Antigen-Specific Immunological Tolerance

This study shows that synthetic glycosylation can effectively target antigens to hepatic and lymph node antigen-presenting cells, leading to local and systemic tolerogenic responses. Additionally, presentation of antigen released from the glycoconjugate is mainly mediated by lymph node-resident dendritic cells, highlighting the importance of targeting antigen to specific cellular components for inducing tolerance.
