Dental stem cell-derived extracellular vesicles as promising therapeutic agents in the treatment of diseases

Ye Li et al.

Summary: Dental stem cells (DSCs) are an important source of mesenchymal stem cells (MSCs) and have been used for the treatment of various diseases. The therapeutic effects of DSCs are attributed to their extracellular vesicles (DSC-EVs), which carry bioactive contents and have therapeutic potential. This review provides insights into the therapeutic effects of DSC-EVs and highlights challenges and future directions for their clinical translation.

The impact of storage on extracellular vesicles: A systematic study

Stefano Gelibter et al.

Summary: Storage has a significant impact on extracellular vesicles (EVs) samples, leading to a decrease in concentration, reduction in purity, and generation of artefactual particles. It is recommended to process fresh, non-archival samples for EVs analysis, depending on downstream analyses and experimental settings.

Extracellular vesicles derived from mesenchymal stem/stromal cells: The regenerative impact in liver diseases

Adriana Psaraki et al.

Summary: Liver dysfunctions are often treated with liver transplantation, but a new concept in therapy involves using extracellular vesicles secreted by mesenchymal stem cells to overcome limitations and promote liver repair. Studies have shown that these vesicles have the potential to ameliorate systemic inflammation and positively impact various pathways associated with liver diseases.

Effects of BMSC-Derived EVs on Bone Metabolism

Xuchang Zhou et al.

Summary: Extracellular vesicles (EVs) are small membrane vesicles that can be secreted by most cells. EVs, especially BMSC-EVs, have therapeutic effects such as tissue repair, inhibition of macrophage polarization, and promotion of angiogenesis. BMSC-EVs also serve as efficient nanocarriers for drug delivery with advantages such as low immunogenicity, no ethical controversy, good stability, and easy storage.

Dental stem cell-derived extracellular vesicles transfer miR-330-5p to treat traumatic brain injury by regulating microglia polarization

Ye Li et al.

Summary: This study reveals the potential therapeutic effect of extracellular vesicles (EVs) secreted by stem cells from human exfoliated deciduous teeth (SHED) in treating traumatic brain injury (TBI). By transferring miR-330-5p, SHED-EVs can regulate microglia polarization and alleviate inflammation, thus promoting functional recovery in TBI.

Functional heterogeneity of mesenchymal stem cells from natural niches to culture conditions: implications for further clinical uses

Luis A. Costa et al.

Summary: Studies have shown that mesenchymal stem cells (MSC) have therapeutic potential in various organs and tissues. However, their functional heterogeneity poses a challenge in transferring their capabilities to clinical applications. Proper selection of MSC based on their functional potential and improving their features in the artificial niche are essential for personalized and precise medicine.

Regulation of exosome production and cargo sorting

Hong Wei et al.

Summary: Cellular communication through the exchange of biological information, mainly in the form of proteins and RNAs, is mediated by extracellular vesicles like exosomes. Understanding the molecular mechanisms of exosome secretion and cargo sorting is crucial for shedding light on their physiological and pathological functions, as well as for exploring potential therapeutic opportunities in clinical applications.

Exosomal miR-125b-5p deriving from mesenchymal stem cells promotes tubular repair by suppression of p53 in ischemic acute kidney injury

Jing-Yuan Cao et al.

Summary: In this study, MSC-exos were found to preferentially target injured kidneys and localize to proximal tubules in ischemic AKI. The researchers demonstrated that MSC-exos ameliorated ischemic AKI and promoted tubular repair by targeting the cell cycle arrest and apoptosis of TECs through the miR-125b-5p/p53 pathway. This study provides new insights into the role of MSC-exos in renal tubule repair and highlights their potential as a promising therapeutic strategy for AKI.
