Substrate Sequence Controls Regioselectivity of Lanthionine Formation by ProcM

Tung Le et al.

Summary: Lanthipeptides, a type of ribosomally synthesized and post-translationally modified peptides, are mostly matured from single precursor peptides in bacteria, while cyanobacteria have the capability to combinatorially synthesize a variety of lanthipeptides from multiple substrate peptides. The study found that lanthionine formation is determined by substrate sequences rather than the enzyme itself, and demonstrated a novel method for separating lanthipeptide constitutional isomers efficiently. The research provides insights into factors influencing cyclization outcomes and suggests potential for deep learning approaches in predicting ring patterns of combinatorial biosynthesis products.

Functional Expression and Characterization of the Highly Promiscuous Lanthipeptide Synthetase SyncM, Enabling the Production of Lanthipeptides with a Broad Range of Ring Topologies

Patricia Arias-Orozco et al.

Summary: Lanthipeptides are ribosomally synthesized and post-translationally modified peptides characterized by stability and functionality provided by lanthionine rings. Heterologous expression of lanthipeptide biosynthetic enzymes offers a system for designing and producing new lanthipeptides with biotechnological or clinical properties. SyncM from marine cyanobacteria has been identified as a promiscuous enzyme with relaxed substrate specificity for producing diverse lanthipeptides.

Nisin- and Ripcin-Derived Hybrid Lanthipeptides Display Selective Antimicrobial Activity against Staphylococcus aureus

Xinghong Zhao et al.

Summary: This study demonstrates the synthesis of novel macrocyclic lanthipeptides using thanatin and rip-thanatin as templates, showing selective antimicrobial activity against S. aureus, including an antibiotic-resistant MRSA strain. The hybrid peptides of nisin(1-20) and ripcin, ripcin B-G, which are inactive against Gram-negative pathogens, exhibited substantial antimicrobial activity against tested Gram-negative pathogens and were highly resistant against the nisin resistance protein NSR.

Mechanisms and Evolution of Diversity-Generating RiPP Biosynthesis

Tung Le et al.

Summary: This review discusses the prochlorosin and cyanobactin biosynthetic pathways in cyanobacteria, which are examples of pathways that generate diversity. It explores the post-translational modifications catalyzed by a single enzyme and the substrate-tolerant enzymes that further diversify the peptide products. Possible evolutionary models to explain the enzymatic tolerance of these systems are also discussed.
