Increased susceptibility to organic dust exposure-induced inflammatory lung disease with enhanced rheumatoid arthritis-associated autoantigen expression in HLA-DR4 transgenic mice

Jill A. Poole et al.

Summary: Immunogenetic factors, environmental and occupational exposures are associated with the development of rheumatoid arthritis (RA), RA-associated lung disease, and other primary lung disorders. Inhalants can trigger the expression of lung autoantigens, leading to inflammatory and fibrotic immune responses. In this study, the authors used an established model of organic dust extract (ODE) exposure in mice to investigate the gene-environment interactions contributing to lung inflammation. They found that ODE exposure resulted in increased lung infiltration, proinflammatory cytokine levels, and ectopic lymphoid aggregates in HLA-DR4 transgenic mice. ODE also induced protein modifications and increased the expression of autoantibodies. These findings suggest that environmental insults can enhance lung inflammation and contribute to the development of RA-associated lung disease.

Summary: Airborne biohazards, such as LPS, can exacerbate rheumatoid arthritis and contribute to associated lung diseases. Patients exposed to LPS may be more susceptible to developing interstitial lung diseases.

Summary: Through single cell RNA sequencing and gene expression analysis in a murine model used for studying RA-associated lung disease, it was found that myeloid immune cells such as macrophages, monocytes, and neutrophils play important roles in arthritis induction, which may contribute to reducing disease burden.
