Senescence and the tumor-immune landscape: Implications for cancer immunotherapy

Loretah Chibaya et al.

Summary: Cancer therapies not only affect tumor cells, but also the tumor-immune microenvironment. Induction of cellular senescence can lead to durable tumor cell arrest and remodeling of the tumor-immune microenvironment through the production of SASP. Depending on the therapy, the SASP can either promote anti-tumor immunity or chronic inflammation, affecting the success of the treatment.

Clinical Value of EZH2 in Hepatocellular Carcinoma and Its Potential for Target Therapy

An-Na Bae et al.

Summary: This study investigated the clinical and prognostic value of EZH2 in hepatocellular carcinoma (HCC) and found that EZH2 overexpression is associated with poor prognosis in HCC. Additionally, EZH2 was correlated with N stage and alpha-fetoprotein (AFP) levels in HCC. The study also revealed a negative correlation between EZH2 expression and STAT3 expression.

Prognostic significance of viable tumor size measurement in hepatocellular carcinomas after preoperative locoregional treatment

Yoon Jung Hwang et al.

Summary: The study suggests that residual viable tumor size (VTS) may be more relevant for predicting prognosis in surgically resected hepatocellular carcinoma (HCC) cases, especially in patients who have undergone preoperative locoregional treatment (LRT). Furthermore, VTS > 2 cm was identified as an independent predictive factor for decreased disease-free survival (DFS).

Senescent Tumor Cells Build a Cytokine Shield in Colorectal Cancer

Yong Won Choi et al.

Summary: Senescent tumor cells inhibit infiltration of CD8(+) T cells by secreting CXCL12 and enhancing differentiation of monocytes into M2 macrophages, forming a cytokine barrier to protect nonsenescent tumor cells. Targeting CXCL12 in senescent tumor cells could enhance T cell infiltration and reduce tumor size in CRC, providing a new strategy to overcome immunotherapy resistance.

Hepatocellular carcinoma

Josep M. Llovet et al.

Summary: Liver cancer, particularly hepatocellular carcinoma (HCC), poses a significant global health challenge with rising incidence projected to exceed 1 million cases by 2025. Infection by hepatitis B and C viruses remains a major risk factor for HCC, while non-alcoholic steatohepatitis is emerging as a more common risk factor in Western countries. Advances in systemic therapies for HCC, including immunotherapies and targeted therapies, are expected to revolutionize the management of this disease.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Locoregional therapies in the era of molecular and immune treatments for hepatocellular carcinoma

Josep M. Llovet et al.

Summary: Locoregional therapies, including local ablation and transarterial chemoembolization, play a crucial role in the management of hepatocellular carcinoma. Recent studies have shown potential benefits of combining these local therapies with systemic treatments, such as molecular therapies, to enhance therapeutic effects.

The Persistence of Hepatitis C Virus Infection in Hepatocytes Promotes Hepatocellular Carcinoma Progression by Pro-Inflammatory Interluekin-8 Expression

Ciniso Sylvester Shabangu et al.

Summary: Persistent HCV infection affects the interaction between the virus and host cell, leading to cell death or growth through different signaling pathways. It induces inflammatory responses and alters cell adhesion and chemokine signaling, promoting hepatocarcinogenesis.

Reprogramming lipid metabolism prevents effector T cell senescence and enhances tumor immunotherapy

Xia Liu et al.

Summary: The functional state of T cells is critical for effective antitumor immunity. T cell senescence driven by malignant tumor cells and regulatory T cells is a common feature in cancers, characterized by unbalanced lipid metabolism and accumulation of lipid droplets. Inhibition of group IVA phospholipase A(2) can reprogram effector T cell lipid metabolism, prevent T cell senescence, and enhance antitumor immunity and immunotherapy efficacy.
