Time to Consider the Exposome Hypothesis in the Development of the Obesity Pandemic

Victoria Catalan et al.

Summary: The main causes of the obesity epidemic are genetics and overnutrition, along with a decline in physical activity. However, there may be other factors, referred to as the exposome, that contribute to obesity. Broad interventions are needed to better identify and understand these factors.

Inhibition of CXXC5 function reverses obesity-related metabolic diseases

Seol Hwa Seo et al.

Summary: The overexpression of CXXC5 and suppression of Wnt/β-catenin signaling in visceral adipose tissues of obesity-related diabetes patients have been identified. By interfering with the binding between CXXC5 and Dvl, the inhibition of CXXC5 function can restore the Wnt/β-catenin signaling and reverse metabolic abnormalities, providing a potential therapeutic strategy for obesity-related diabetes.

Metabolic improvement and liver regeneration by inhibiting CXXC5 function for non-alcoholic steatohepatitis treatment

Seol Hwa Seo et al.

Summary: This study found that the small molecule KY19334 can effectively treat obesity-related liver disease NASH by activating the Wnt/β-catenin signaling pathway. The treatment suppressed inflammation, steatosis, oxidative stress, and cell death, reversed liver fibrosis, and enhanced liver regeneration.

Global pandemics interconnected - obesity, impaired metabolic health and COVID-19

Norbert Stefan et al.

Summary: Obesity and impaired metabolic health are established risk factors for NCDs such as type 2 diabetes mellitus, cardiovascular disease, neurodegenerative diseases, cancer, and MAFLD, and have been linked to an increased risk of severe COVID-19. Specifically, visceral obesity and characteristics of impaired metabolic health are associated with a high risk of severe COVID-19.

The Role of TCF7L2 in Type 2 Diabetes

Laura del Bosque-Plata et al.

Summary: TCF7L2 is the most potent locus for type 2 diabetes (T2D) risk and is consistently associated with the disease across diverse populations with different genetic origins. The exact mechanisms by which TCF7L2 affects T2D are still not well understood.
