Focus on Molecular Functions of Anti-Aging Deacetylase SIRT3

Jarmila Nahalkova

Summary: SIRT3 plays an important role in maintaining mitochondrial integrity and has reversible effects on cellular aging. Its functions include involvement in thermogenesis, neurodegenerative diseases, and non-alcoholic fatty liver disease. The main regulatory nodes within its interaction network are subunits of the complex I of the mitochondrial respiratory chain. SIRT3 also acts as a repressor of BACE1 through a specific pathway, suggesting it as a potential therapeutic target for Alzheimer's disease.

Peripheral and cognitive benefits of physical exercise in a mouse model of midlife metabolic syndrome

Farida El Gaamouch et al.

Summary: This study investigates the impact of high fat diet on the development of metabolic syndrome in middle-aged mice and the potential benefits of voluntary physical exercise on peripheral and brain cognitive function. The results show that metabolic syndrome impairs cognitive function and is associated with gene dysregulation in metabolic pathways. Eight-week voluntary wheel running improves glucose control and enhances learning and memory, as well as restores gene expression involved in energy metabolism in the brain. These findings suggest that voluntary physical exercise can be a recommended therapeutic intervention for metabolic syndrome and associated diseases.

Synaptic basis of Alzheimer's disease: Focus on synaptic amyloid beta, P-tau and mitochondria

Albin John et al.

Summary: Alzheimer's disease is a progressive disease associated with synaptic dysfunction, with researchers striving to find more information on disease progression and pathogenesis. Synaptic pathology and mitochondrial oxidative damage are early events in the progression of AD, with synaptic loss and damage being key correlates of cognitive deficits in patients. As the disease advances, significant changes occur at the synapse, shedding light on the disease progression and serving as potential markers and therapeutic targets.

Oxidative Damage and Antioxidant Response in Frontal Cortex of Demented and Nondemented Individuals with Alzheimer's Neuropathology

Anna Fracassi et al.

Summary: The correlation between the pathological features of AD and dementia has been challenged by the presence of individuals with AD neuropathology but intact cognitive function. It suggests that unknown mechanisms may be triggered to resist the detrimental effects of Aβ accumulation. The study found that AD brains exhibit a dramatic redox imbalance, while NDAN individuals show low oxidative damage, possibly due to a lack of PGC1 alpha miRNA-485-related inhibition.

NAD plus supplementation reduces neuroinflammation and cell senescence in a transgenic mouse model of Alzheimer's disease via cGAS-STING

Yujun Hou et al.

Summary: Research reveals that levels of NAD* decrease, and inflammation increases in the brains of Alzheimer's disease patients, while treatment with NR can increase brain NAD* levels, reduce inflammation, and improve cellular senescence. NR treatment is beneficial for neuroinflammation and cellular senescence through a cGAS-STING dependent pathway, inducing mitophagy and improving cognitive function.

Effects of Fasting and Feeding on Transcriptional and Posttranscriptional Regulation of Insulin-Degrading Enzyme in Mice

Carlos M. Gonzalez-Casimiro et al.

Summary: This study investigates the regulation of insulin-degrading enzyme (IDE) in the liver, skeletal muscle, and kidneys of mice under fasting and refeeding conditions, revealing a direct correlation between circulating lactate concentrations and IDE activity and protein levels. The study also suggests that IDE regulation in liver and muscle tissues is more complex than previously expected, with fasting/refeeding having little influence on renal IDE regulation.
