Vaping and Lung Inflammation and Injury

Jin-Ah Park et al.

Summary: The use of e-cigarettes, initially seen as a solution to quit conventional smoking, is paradoxically growing rapidly among nonsmokers and leading to adverse health effects such as acute lung injury. The mechanisms behind this novel disease, EVALI, are still unknown, hindering the development of biomarkers and therapeutics. Furthermore, chronic e-cigarette exposure has been linked to chronic lung diseases and systemic effects on multiple organs.

Summary: This study found that vaping electronic cigarettes may cause cardiopulmonary injury and inflammation, with more severe effects when heating is involved. The roles of CARD9 and NLRP3 in this process were identified.

Tenascin-C expression in the lymph node pre-metastatic niche in muscle-invasive bladder cancer

Christopher R. Silvers et al.

Summary: Elevated expression of tenascin-C in regional lymph nodes may be a predictor of bladder cancer metastasis and associated with worse survival. Urinary extracellular vesicles contain cytokines that are positively correlated with lymph node tenascin-C expression. Bladder cancer extracellular vesicles induce tenascin-C expression in fibroblasts in an NF-kappa B-dependent manner.

Non-stem bladder cancer cell-derived extracellular vesicles promote cancer stem cell survival in response to chemotherapy

Wei-Min Chung et al.

Summary: This study revealed that EVs derived from NSCCs contain cargo proteins enriched in proteostasis-related functions, significantly impacting the development of CSCs to be more intrinsically chemoresistant and aggressive. This novel communication mechanism between NSCC-EVs and CSCs may promote disease progression and the acquisition of chemoresistance.

HSP47 contributes to angiogenesis by induction of CCL2 in bladder cancer

Wenlong Ma et al.

Summary: The study revealed that HSP47 is overexpressed in bladder cancer and is associated with poor prognosis. Down-regulation of HSP47 can suppress angiogenesis through activation of the ERK pathway and induction of CCL2. This highlights HSP47 as a potential anti-angiogenesis target for bladder cancer therapy.

p97/VCP inhibition causes excessive MRE11-dependent DNA end resection promoting cell killing after ionizing radiation

Susan Kilgas et al.

Summary: The ATPase p97 interacts with the MRN complex on chromatin and inactivation of p97 affects DNA repair and radiosensitivity. Inhibition of p97 function increases tumor cell killing following ionizing radiation by promoting MRE11 nuclease accumulation, suggesting potential use of p97 inhibitors in radiotherapy patients.
