How to Couple LC-IRMS with HRMS-A Proof-of-Concept Study

Robert G. H. Marks et al.

Summary: Compound-specific stable isotope analysis (CSIA) is a valuable technique for determining small variations in isotopes in complex mixtures, but may lose structural information. The combination of LC with IRMS and HRMS allows for direct identification of unknown species, but technical challenges must be overcome.

Tracing the Trophic Fate of Aquafeed Macronutrients With Carbon Isotope Ratios of Amino Acids

Thomas Larsen et al.

Summary: To meet future seafood demands, researchers are exploring the use of ingredients derived from algae and other sustainable sources as replacers for traditional aquafeed ingredients. Algal ingredients are being promoted for their sustainability and additional functional attributes in farmed aquatic animals. Stable carbon isotope analysis of single amino acids (AAs) shows promise as a nutritional biomarker in aquaculture to understand the digestion and metabolism of algal macronutrients. Although there have been few applications to date, there is potential for further development and implementation of delta C-13(AA)-based approaches to improve understanding of protein origins and macronutrient metabolic routing.

Insights into amino acid fractionation and incorporation by compound-specific carbon isotope analysis of three-spined sticklebacks

Tobias Hesse et al.

Summary: Interpretation of stable isotope data is crucial in ecology for studying animal diets, migration patterns, and physiology. This study analyzed the delta C-13 values of individual amino acids in muscle and liver tissue of three-spined sticklebacks on a high protein diet. The results showed fast response of liver amino acids to small shifts in dietary stable isotope compositions. Trophic fractionation of specific amino acids was observed and discussed, suggesting the potential use of certain amino acids as dietary proxies.

Climate change facilitates a parasite's host exploitation via temperature-mediated immunometabolic processes

Jorn P. Scharsack et al.

Summary: Global climate change can influence the interaction between hosts and parasites, with rising temperatures potentially exacerbating host exploitation by parasites. The study showed that warm temperatures enhanced parasite growth and infectivity to final hosts, while hosts exhibited lower body condition and less active immune system at high temperatures. The data suggests that parasite exposure combined with rising temperatures can shift the host's immunometabolism, providing nutrients for parasite growth and suppressing immune defense.

Zone of Interaction Between the Parasite and the Host: Protein Profile of the Body Cavity Fluid of Gasterosteus aculeatus L. Infected with the Cestode Schistocephalus solidus (Muller, 1776)

Albina Kochneva et al.

Summary: This study aimed to describe the excretory-secretory proteins from the helminth Schistocephalus solidus and its intermediate host, the three-spined stickleback Gasterosteus aculeatus L. Mass-spectrometry analysis revealed 215 identified proteins, with infected fish and helminths showing a predominance of components from the immune defense system in their washes.

Stable nitrogen isotope analysis of amino acids as a new tool to clarify complex parasite-host interactions within food webs

Philip M. Riekenberg et al.

Summary: Traditional bulk isotopic analysis is limited in describing parasite-host relationships in food webs, while compound-specific stable isotopes from amino acids offer a promising novel approach for mapping these interactions. The compound-specific method provided clearer trophic discrimination factors and accurately identified parasitic trophic interactions, highlighting its usefulness in clarifying feeding strategies for parasitic species within marine food webs. Despite the limitation of applying a universal trophic discrimination factor, compound-specific stable isotope analysis remains a valuable tool to increase understanding of parasitic interactions.

The parasite Schistocephalus solidus secretes proteins with putative host manipulation functions

Chloe Suzanne Berger et al.

Summary: The study used proteomics to characterize the secretome of Schistocephalus solidus, a manipulative cestode in its obligatory intermediate fish host. The results showed the presence of molecules with putative host manipulation functions, many of which are species-specific.
