The bitter end: T2R bitter receptor agonists elevate nuclear calcium and induce apoptosis in non-ciliated airway epithelial cells

Summary: Bitter taste receptors (T2Rs) located on airway motile cilia and nucleus play a crucial role in regulating innate immune responses and apoptosis in airway diseases and cancer. T2R agonists selectively increase nuclear and mitochondrial calcium through a G-protein-coupled receptor phospholipase C mechanism, leading to nitric oxide production and apoptosis in cells. Targeting T2Rs may provide a potential therapeutic strategy for treating airway diseases and activating cancer cell apoptosis.

Summary: T2R bitter taste receptors in airway motile cilia increase ciliary beat frequency and nitric oxide production. The expression of T2Rs is correlated with the differentiation of nasal epithelial cells, but not affected by cystic fibrosis. Diphenhydramine, as a T2R agonist, increases nitric oxide production and ciliary beat frequency, while inhibiting the growth and biofilm formation of Pseudomonas aeruginosa. Therefore, diphenhydramine-derived compounds may have potential clinical usefulness in CF-related chronic rhinosinusitis as a topical therapy.

This study shows that SARS-CoV-2 infection leads to rapid loss of airway motile cilia, affecting cilia clearance function, with reduced expression of the ciliogenesis regulator Foxj1 preceding cilia loss. Further analysis in Syrian hamsters confirms the loss of motile cilia in vivo.
