Baicalein ameliorates ulcerative colitis by improving intestinal epithelial barrier via AhR/IL-22 pathway in ILC3s

Yan-yang Li et al.

Summary: This study elucidates the molecular mechanisms underlying the protective effect of baicalein on intestinal barrier function in colitis mice, demonstrating that baicalein improves gut inflammation, reduces intestinal permeability, and restores tight junctions possibly via promoting AhR/IL-22 pathway. Co-administration of AhR antagonist CH223191 partially blocks the therapeutic effects of baicalein, while AhR agonist FICZ ameliorates symptoms and gut barrier function in colitis mice.

CCN1 interacts with integrins to regulate intestinal stem cell proliferation and differentiation

Jong Hoon Won et al.

Summary: This study reveals that the matricellular protein CCN1 is a niche factor for intestinal stem cells, regulating their proliferation and differentiation through integrin signaling. CCN1 interacts with integrins alpha(v)beta(3)/alpha(v)beta(5) to activate distinct downstream pathways, leading to Notch activation for differentiation via NF-kappaB and Wnt signaling control for proliferation via Src-mediated YAP activation and Dkk1 expression. Moreover, CCN1 and YAP amplify each other's activities in a regulatory loop.

Optimized human intestinal organoid model reveals interleukin-22-dependency of paneth cell formation

Gui-Wei He et al.

Summary: IL-22 has opposing roles in intestinal pathophysiology, slowing the growth of small intestinal organoids while promoting the formation of Paneth cells and the expression of host defense genes.

Inflammation triggers ILC3 patrolling of the intestinal barrier

Angelique Jarade et al.

Summary: Serafini and colleagues find that intestinal villus ILC3s acquire migratory 'patrolling' attributes and enhanced cytokine expression in response to inflammation. These findings reveal notable differences between ILC3s and T cells in immunosurveillance, with ILC3s showing adaptation towards mucosal immunosurveillance after inflammation.

Fucose Ameliorates Tritrichomonas sp.-Associated Illness in Antibiotic-Treated Muc2-/- Mice

Kseniya M. Achasova et al.

Summary: This study found that Muc2(-/-) mice harboring a gut protozoan infection Tritrichomonas sp. are susceptible to antibiotic-induced depletion of bacterial microbiota. Effective suppression of the protozoan infection with metronidazole or L-fucose administration can ameliorate illnesses observed in antibiotic-treated Muc2(-/-) mice. Further investigation of the role of fucose in protozoan adhesion to host cells may be valuable.

Mechanisms and regulation of IL-22-mediated intestinal epithelial homeostasis and repair

Lori Patnaude et al.

Summary: The study found that IL-22 promotes expansion and proliferation of intestinal epithelial stem cells, as well as production of anti-microbial peptides. Additionally, IL-22 alters the composition of the mucus layer. However, the IL-22-associated epithelial phenotype differs in the presence of butyrate and immune cells.

AHR in the intestinal microenvironment: safeguarding barrier function

Brigitta Stockinger et al.

Summary: The Aryl hydrocarbon receptor (AHR) signaling pathway plays significant roles in the intestine, regulating various cell types and impacting inflammation and tumorigenesis. AHR is a ligand-dependent transcription factor that translocates to the nucleus upon ligand binding, where it interacts with other proteins to modulate gene expression. Research has shown that AHR not only responds to environmental pollutants, but also serves important beneficial functions in immune, epithelial, endothelial, and neuronal cells within the intestinal microenvironment.
