Statistical power in vitamin D randomized control trials investigating biomarkers as continuous outcomes

Summary: This study investigated the power and sample size requirements in vitamin D supplementation randomized controlled trials (RCTs) and found that the increase in 25OHD in the intervention arm and population heterogeneity substantially impact the power of the trials. Sample size determination through simulation was shown to be a powerful approach for non-standard trials and can be easily applied to other intervention-outcome pairs.

Summary: This study found that low winter ambient UVB and prolonged vitamin D deficiency are associated with an increased risk of AAV relapse across all phenotypes. However, the development of a granulomatous phenotype does not appear to be directly mediated by vitamin D. Further research is needed to determine if sufficient vitamin D status could reduce relapse propensity in AAV.

An observational and Mendelian randomisation study on vitamin D and COVID-19 risk in UK Biobank

Summary: While no significant association between vitamin D levels and COVID-19 infection risk was found, a strong inverse relationship was observed between ambient UVB radiation and COVID-19 hospitalization and death rates. Additionally, genetically-predicted vitamin D levels showed a greater protective effect in stronger ambient UVB radiation. Further studies are needed to confirm the potential benefits of vitamin D supplementation in protecting against severe COVID-19 outcomes.
