Insulin Resistance: From Mechanisms to Therapeutic Strategies

Shin-Hae Lee et al.

Summary: Insulin resistance, characterized by reduced responsiveness of insulin-targeting tissues to physiological levels of insulin, is a key pathogenic component of many metabolic diseases. This review summarizes the functions of insulin in glucose metabolism and discusses the mechanisms proposed for insulin resistance, such as ectopic lipid accumulation, endoplasmic reticulum stress, and inflammation. Potential therapeutic strategies for addressing insulin resistance are also suggested.

Insulin regulates arginine-stimulated insulin secretion in humans

Florencia Halperin et al.

Summary: This study investigates the regulatory role of insulin in the beta cell response to arginine stimulation in healthy humans. The results demonstrate that pre-exposure to insulin enhances arginine-stimulated insulin secretion. This finding reveals the physiological role of insulin in regulating beta cell response to non-glucose secretagogues.

High-Throughput Analysis of Amino Acids for Protein Quantification in Plant and Animal-Derived Samples Using High Resolution Mass Spectrometry

Priyanka Reddy et al.

Summary: A fast LC-MS method for determining amino acids has been developed, requiring only three simple steps of hydrolysis, neutralisation, and sample dilution with a borate buffer solution. The method shows excellent repeatability and reproducibility for amino acid content, peak area, and retention time. Detection levels on column are below 0.1 μM for most amino acids.

Moderate over-feeding of different sources of metabolizable energy and protein improved gestational insulin resistance markers and maternal metabolic status of sheep around lambing

M. Moradi et al.

Summary: This study aimed to investigate the effect of moderate over-nutrition of energy and protein on prepartal insulin resistance, metabolic status, and performance of ewes. The results showed that moderately over-fed ewes with extra protein and energy had improved insulin sensitivity and milk production, as well as better metabolic status around parturition.

Insulin action, type 2 diabetes, and branched-chain amino acids: A two-way street

Phillip J. White et al.

Summary: The association of obesity and insulin resistance with increased levels of branched-chain and aromatic amino acids and decreased glycine levels is well-established. Human metabolomics and genetic studies have further confirmed these observations, with preclinical studies identifying mechanisms involved in amino acid homeostasis disruption and its connection to dysregulated glucose and lipid metabolism, particularly in the development of insulin resistance, type 2 diabetes, and cardiovascular diseases.

Insulin resistance, cardiovascular stiffening and cardiovascular disease

Summary: Insulin resistance is typically accompanied by metabolic insulin resistance and cardiovascular disease, including the correlation of reduced nitric oxide production leading to increased blood flow. Insulin resistance and hyperinsulinemia promote cardiovascular diseases by increasing vascular stiffness.

Associations between insulin resistance and adverse pregnancy outcomes in women with gestational diabetes mellitus: a retrospective study

Jing Lin et al.

Summary: This study found that IR in the second trimester significantly increased the risk of hypertensive disorders of pregnancy and large for gestational age in women with GDM, while other adverse pregnancy outcomes were not significantly increased. Additionally, factors such as FPG, HbA1c, TG in early pregnancy, pre-pregnant BMI, and weight gain before GDM diagnosis were all identified as independent risk factors for IR.
