Characteristics of an In Vitro Mesenteric Lymph Node Cell Suspension Model and Its Possible Association with In Vivo Functional Evaluation

Saisai Feng et al.

Summary: This study investigated the immune-responsive patterns of gut microbes using an in vitro MLN model and compared the functional differences between different response patterns in an in vivo TNBS-challenge model. The results showed that different strains of gut microbes evoke immune responses in different ways in vitro and that a mixture of two strong immunogenic strains increases mortality in mice. Further research is needed to determine the underlying mechanisms.

A. muciniphila Supplementation in Mice during Pregnancy and Lactation Affects the Maternal Intestinal Microenvironment

Yuli Qi et al.

Summary: This study investigated the impact of Akkermansia muciniphila (A. muciniphila) supplementation during pregnancy and lactation on maternal and offspring health. The results showed that dams fed with A. muciniphila had enhanced intestinal barrier, altered gut microbiota composition and diversity, and significant enrichment of A. muciniphila and Ruminococcus_1 in offspring. However, fecal metabolites only changed slightly in the A. muciniphila group, and the abundance of A. muciniphila in the fecal microbiota of offspring did not significantly increase. The fecal metabolic profile of three-week-old offspring mice fed with A. muciniphila was significantly changed. These findings suggest that A. muciniphila supplementation during pregnancy and lactation is safe and may have a more beneficial effect on dams.

Management of Crohn Disease A Review

Kelly Cushing et al.

Summary: Crohn disease is a chronic gastrointestinal inflammatory disease that requires early and effective control of inflammation. Treatment options include medication, surgery, and vaccination for infection prevention. Physicians should consider disease severity, patient risk stratification, age, and other clinical factors when determining the best treatment strategy.

Bifidobacterium longum Ameliorates Dextran Sulfate Sodium-Induced Colitis by Producing Conjugated Linoleic Acid, Protecting Intestinal Mechanical Barrier, Restoring Unbalanced Gut Microbiota, and Regulating the Toll-Like Receptor-4/Nuclear Factor-κB Signaling Pathway

Yang Chen et al.

Summary: Bifidobacterium longum CCFM681 alleviated colitis by protecting the intestinal mechanical barrier, modulating gut microbiota, inhibiting the TLR4/NF-kappa B pathway, and increasing colonic CLA concentrations. The effectiveness of relieving colitis showed a significantly positive correlation with CLA concentrations.

Microbial and metabolic features associated with outcome of infliximab therapy in pediatric Crohn's disease

Yizhong Wang et al.

Summary: The study revealed that pediatric CD patients have gut microbial dysbiosis and altered metabolism after IFX therapy, which is related to treatment outcomes. The effects of IFX treatment may be mediated by enriching bacteria taxa that produce SCFAs and BSH, thereby inhibiting inflammation and restoring bile acid metabolism. Some fecal bacteria and metabolites may serve as predictors of IFX therapy outcomes for pediatric CD patients.

Soluble Fraction from Lysate of a High Concentration Multi-Strain Probiotic Formulation Inhibits TGF-β1-Induced Intestinal Fibrosis on CCD-18Co Cells

Francesca Lombardi et al.

Summary: This study explores the anti-fibrotic impact of two high concentration multi-strain probiotic formulations on TGF-beta 1-activated human intestinal colonic myofibroblast CCD-18Co. The results suggest that the soluble fraction of Vivomixx(R) formulation, but not VSL#3(R), can antagonize the pro-fibrotic effects of TGF-beta 1 on CCD-18Co cells, preventing all cellular and molecular parameters related to fibrotic phenotype. The differences observed between the two probiotics used in the fibrosis model may be due to the different number of strains in different proportions.

Summary: The study found that IL-22 promotes expansion and proliferation of intestinal epithelial stem cells, as well as production of anti-microbial peptides. Additionally, IL-22 alters the composition of the mucus layer. However, the IL-22-associated epithelial phenotype differs in the presence of butyrate and immune cells.

A bacterial bile acid metabolite modulates Treg activity through the nuclear hormone receptor NR4A1

Wei Li et al.

Summary: Bile acids regulate immune balance by affecting T cell differentiation, with isoalloLCA promoting T-reg cell differentiation through chromatin structure changes; gut bacteria capable of producing isoalloLCA belong to common human gut bacterial phylum Bacteroidetes, and NR4A1 is essential for isoalloLCA's effects on T-reg cells; significantly reduced levels of isoalloLCA in patients with inflammatory bowel diseases suggest a critical role for isoalloLCA and its bacterial producers in maintaining immune homeostasis in humans.

Bifidobacterium bifidum Enhances the Intestinal Epithelial Tight Junction Barrier and Protects against Intestinal Inflammation by Targeting the Toll-like Receptor-2 Pathway in an NF-κB-Independent Manner

Rana Al-Sadi et al.

Summary: The probiotic bacteria BB1 causes a strain-specific enhancement of the intestinal TJ barrier through a novel mechanism involving BB1 attachment to the enterocyte TLR-2 receptor complex and activation of p38 kinase pathway. This enhancement is sustained, and BB1 strain designated as BB1 causes a maximal enhancement in TJ barrier function. The mechanism of BB1 enhancement of intestinal TJ barrier is mediated by the p38 kinase pathway, not the NF-kappa B signaling pathway.
