Thermoadaptation in an Ancestral Diterpene Cyclase by Altered Loop Stability

David A. Hueting et al.

Summary: In this paper, a hyperstable variant of the terpene cyclase PtmT2 was created using ancestral sequence reconstruction, and molecular dynamics simulations were performed to investigate the molecular mechanisms contributing to activity at an elevated temperature and increased melting temperature. The study highlights the importance of loop flexibility in enzyme thermoadaptation.

A New Thermophilic Ene-Reductase from the Filamentous Anoxygenic Phototrophic Bacterium Chloroflexus aggregans

Marina Simona Robescu et al.

Summary: The study investigated a new thermophilic ene-reductase homologue, CaOYE, from the photosynthetic extremophile microorganism Chloroflexus aggregans, showing high potential as a robust biocatalyst with high thermostability and good solvent tolerance. Although it displayed a limited substrate spectrum, further investigations could reveal peculiar chemical activities due to the metabolic ductility and versatility of Chloroflexus aggregans. The structural and functional characterization of CaOYE provides new insights for the development and application of this class of enzymes.

Catalytic mechanism of ancestral L-lysine oxidase assigned by sequence data mining

Sayaka Sugiura et al.

Summary: In this study, 28 original sequences were assigned to uncharacterized enzymes in the FAD-dependent oxidase family expressed in certain species of bacteria. The enzyme named AncLLysO was shown to exhibit L-lysine oxidase activity. Through crystal structure determination, it was found that AncLLysO recognizes L-Lys by hydrogen bond formation with specific residues and undergoes structural changes upon substrate binding. The study demonstrates the effectiveness of combining database sequence classification and ancestral sequence reconstruction in discovering new enzymes and elucidating their functions.

Ancestral sequence reconstruction - An underused approach to understand the evolution of gene function in plants?

Federico Scossa et al.

Summary: Ancestral proteins were more promiscuous than modern ones, with specificity often evolving after gene duplication; some modern proteins are found to have evolved de novo from ancestors lacking those functions; new interactions evolved from just a few mutations, suggesting acquisition of new functions is not difficult but often lost to subsequent mutations.
