KIR+CD8+ T cells suppress pathogenic T cells and are active in autoimmune diseases and COVID-19

Jing Li et al.

Summary: The study reveals that CD8(+) T cells expressing inhibitory receptors play a regulatory role in immune responses in both humans and mice, and their increased presence is observed in various autoimmune diseases and COVID-19 patients. The findings suggest that these regulatory CD8(+) T cells are involved in suppressing pathogenic T cells in autoimmune and infectious diseases.

Granzyme K+ CD8 T cells form a core population in inflamed human tissue

A. Helena Jonsson et al.

Summary: CD8 T cells play a significant role in the pathogenesis of rheumatoid arthritis by producing pro-inflammatory cytokines. These CD8 T cells are abundant in synovial tissue and belong to an effector population characterized by high expression of granzyme K and low expression of granzyme B and perforin. They are clonally expanded in synovial tissues and maintain their characteristics in blood, suggesting their enrichment in tissues. They also exist in other tissues and diseases, indicating their role as tissue-associated T cells.

Arthritis flares mediated by tissue-resident memory T cells in the joint

Margaret H. Chang et al.

Summary: The recurrence of rheumatoid arthritis is mediated by long-lived synovial resident memory T cells (T-RM) activated by antigen, leading to the recruitment of circulating effector cells. Depletion of T-RM can ameliorate recurrence in a site-specific manner. Human rheumatoid arthritis joint tissues contain a CD8+-predominant T-RM population.

Integrated analysis of multimodal single-cell data

Yuhan Hao et al.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.
