Identification of a subset of immunosuppressive P2RX1-negative neutrophils in pancreatic cancer liver metastasis

Xu Wang et al.

Summary: A subset of immunosuppressive P2RX1-negative neutrophils accumulate in the liver metastases of pancreatic ductal adenocarcinoma (PDAC), expressing increased levels of immunosuppressive molecules such as PD-L1 and having enhanced mitochondrial metabolism. The upregulation of the transcription factor Nrf2 in P2RX1-deficient neutrophils is associated with PD-L1 expression and metabolic reprogramming, contributing to the evasion of antitumor immunity by metastatic PDAC tumors.

Wnt-β-Catenin Signaling in Human Dendritic Cells Mediates Regulatory T-Cell Responses to Fungi via the PD-L1 Pathway

Anupama Karnam et al.

Summary: Interaction between dendritic cells (DCs) and fungi activates the Wnt/beta-catenin pathway in human DCs, promoting immunosuppressive Treg responses. Inhibition of this pathway selectively suppresses Treg responses without affecting the Th1 response, offering a potential therapeutic target against invasive fungal diseases such as those caused by Aspergillus fumigatus.

PD-L1+ neutrophils contribute to injury-induced infection susceptibility

Ajitha Thanabalasuriar et al.

Summary: The rapid increase in neutrophil numbers in the lungs following injury is associated with elevated levels of PD-L1. Neutrophils transiently arrest in the lung vasculature and interact with PD-1 on lung endothelial cells, contributing to infection susceptibility. Treatment with an anti-PD-L1 antibody prevents neutrophil accumulation in the lung, reduces susceptibility to infection, and promotes skin healing.

Tumor-derived lactate inhibit the efficacy of lenvatinib through regulating PD-L1 expression on neutrophil in hepatocellular carcinoma

Haijing Deng et al.

Summary: Neutrophils play a controversial role in tumor development, with PD-L1(+) neutrophils inhibiting the cytotoxicity of anti-tumor immunity. Lenvatinib was found to induce CXCL2 and CXCL5 secretion, promoting recruitment of neutrophils in the tumor microenvironment. Inhibiting PD-L1(+) neutrophils with a COX-2 inhibitor could enhance the antitumor effect of lenvatinib by restoring T cell cytotoxicity.

G-CSF - A double edge sword in neutrophil mediated immunity

Katherine R. Martin et al.

Summary: Neutrophils play a vital role in the host defense against pathogens, and their function is tightly regulated by factors such as G-CSF to prevent tissue damage from excessive response. Therefore, in inflammatory and infectious diseases, neutrophils and the G-CSF pathway may serve as novel therapeutic targets.

Priming with FLO8-deficient Candida albicans induces Th1-biased protective immunity against lethal polymicrobial sepsis

Quan-Zhen Lv et al.

Summary: Preconditioning with the FLO8-deficient mutant of C. albicans provides protection against subsequent lethal infections by inducing Dectin-2/CARD9-mediated IL-10 production. This protection is achieved by preventing C. albicans-induced apoptosis of thymic T cells, leading to enhanced Th1-biased immune responses against fungal infections.

KPNB1-mediated nuclear translocation of PD-L1 promotes non-small cell lung cancer cell proliferation via the Gas6/MerTK signaling pathway

Wenwen Du et al.

Summary: This study demonstrates for the first time that PD-L1 regulates NSCLC cell proliferation through the Gas6/MerTK signaling pathway, and reveals a molecular mechanism by which PD-L1 translocates into the nucleus to promote Gas6 synthesis and activate the MerTK signaling pathway. These findings provide potential implications for PD-L1 targeted immunotherapy in the clinic.
