Deep learning enables genetic analysis of the human thoracic aorta

James P. Pirruccello et al.

Summary: Genome-wide association analyses identified variants associated with thoracic aortic diameter and polygenic score for ascending aortic diameter was correlated with a diagnosis of thoracic aortic aneurysm. Enlargement or aneurysm of the aorta predisposes to dissection, and a deep learning model along with genome-wide association studies successfully identified loci associated with ascending and descending thoracic aortic diameter, highlighting the potential for rapidly defining quantitative traits with deep learning in biomedical images.

A phenome-wide bidirectional Mendelian randomization analysis of atrial fibrillation

Qin Wang et al.

Summary: This study used Mendelian randomization analysis to evaluate the causes and consequences of atrial fibrillation (AF). The results showed that body mass index, height, systolic blood pressure, and genetic liability to coronary artery disease are independent causal risk factors for AF. Additionally, several proteins were identified as having therapeutic potential for AF.

High heritability of ascending aortic diameter and trans-ancestry prediction of thoracic aortic disease

Catherine Tcheandjieu et al.

Summary: This study used automated extraction of ascending aortic diameter from cardiac magnetic resonance images and conducted a genome-wide association study. The polygenic score constructed from multiple loci was able to predict the risk of thoracic aortic aneurysm in different populations. Additionally, the study revealed the causal role of blood pressure in reducing aortic dilation.

Nay Aung et al.

Summary: This study conducted genome-wide association analyses using a deep learning algorithm on cardiovascular magnetic resonance images of UK Biobank participants, and identified 25 unique loci associated with right ventricular structure and function. The findings were validated in a meta-analysis, and revealed associations between RV polygenic risk scores and dilated cardiomyopathy and coronary artery disease.

Summary: This study utilized deep learning models to recognize and measure right heart structures and function. Genome-wide association studies identified several loci associated with right heart measurements, some of which were previously linked with congenital heart disease. Additionally, a polygenic predictor was associated with right ventricular ejection fraction and risk of dilated cardiomyopathy.

Shared genetic pathways contribute to risk of hypertrophic and dilated cardiomyopathies with opposite directions of effect

Rafik Tadros et al.

Summary: The study found a strong genetic correlation between hypertrophic and dilated cardiomyopathies and left ventricular traits, with opposing genetic effects in the two cardiomyopathies. The research also supported a causal association between increased left ventricular contractility and hypertrophic cardiomyopathy risk.

The power of genetic diversity in genome-wide association studies of lipids

Sarah E. Graham et al.

Summary: This study conducted a multi-ancestry, genome-wide genetic discovery meta-analysis of lipid levels in approximately 1.65 million individuals, including 350,000 of non-European ancestries. The findings demonstrate that increasing diversity rather than studying additional individuals of European ancestry results in substantial improvements in fine-mapping functional variants and portability of polygenic prediction, with modest gains in the number of discovered loci and ancestry-specific variants. This highlights the importance of including diverse populations in genetic studies to improve the accuracy and equity of polygenic scores in clinical practice.

Alpha-protein kinase 3 (ALPK3) truncating variants are a cause of autosomal dominant hypertrophic cardiomyopathy

Luis R. Lopes et al.

Summary: The study aimed to determine the frequency of heterozygous truncating ALPK3 variants in patients with hypertrophic cardiomyopathy (HCM) and confirm their pathogenicity through burden testing. The results showed that ALPK3tv carriers had a higher prevalence of apical/concentric patterns of hypertrophy and a short PR interval, indicating a characteristic HCM phenotype. Patients with ALPK3tv also had higher rates of heart failure or cardiac transplantation, with imaging and histopathology revealing extensive myocardial fibrosis and myocyte vacuolation.
