Cryo-EM demonstrates the in vitro proliferation of an ex vivo amyloid fibril morphology by seeding

Summary: In this study, cryo-electron microscopy and other methods were used to analyze the ability of SAA1.1-derived amyloid fibrils to seed solutions of recombinant SAA1.1 protein. The seeded fibrils were found to remodel the main ex vivo fibril morphology and were more stable than unseeded fibrils.

Summary: Systemic AA amyloidosis is a protein misfolding disease caused by the formation of amyloid fibrils from serum amyloid A (SAA) protein. The study shows that amyloid fibrils isolated from mouse tissue differ structurally from in vitro formed fibrils, with the former being more resistant to proteolysis, suggesting that pathogenic amyloid fibrils may originate from proteolytic selection.
