Developmental landscape of human forebrain at a single-cell level identifies early waves of oligodendrogenesis

Summary: The study identified the emergence of the first wave of oligodendrocyte lineage cells in the human forebrain as early as the 8th week of gestation, indicating evolutionary conservation of this process between mice and humans. Through analyzing the spatial expression of 50 key genes, regions in the human ventral fetal forebrain where oligodendrogenesis first occurs were identified.

Developmental Origins of Human Cortical Oligodendrocytes and Astrocytes

Summary: In this study, the researchers integrated analysis of published human cortical single-cell RNA-Seq datasets with immunohistochemical analyses to show that around gestational week 18, EGFR-expressing human cortical truncated radial glial cells give rise to basal multipotent intermediate progenitors. These progenitors go through several rounds of mitosis and generate different types of glial cells and neurons in the developing human cerebral cortex. The findings provide insights into the lineage progression of cortical radial glial cells.

Single-cell atlas of early human brain development highlights heterogeneity of human neuroepithelial cells and early radial glia

Summary: Through single-cell RNA sequencing, nine progenitor populations at the earliest stages of human brain development were identified, including a highly prevalent mesenchymal-like population that disappears once neurogenesis begins. Comparison with mouse progenitor populations revealed two clusters enriched in the early stages of human cortical development. Organoid systems displayed low fidelity to neuroepithelial and early radial glia cell types, but improved as neurogenesis progressed.

Summary: The complexity of human astrocytes in primary tissue requires improved tools for their isolation and molecular characterization. A new immunotagging strategy has been developed to isolate enriched nuclei populations from fresh-frozen human cortex for transcriptome and chromatin sequencing. This protocol is suitable for various human cortical specimens with different pathologies and recommends tissue collection within 24 hours postmortem.
