Anti-centromere antibodies target centromere-kinetochore macrocomplex: a comprehensive autoantigen profiling

Nobuhiko Kajio et al.

Summary: This study reveals that serum autoantibodies target the centromere-kinetochore macrocomplex in patients with SS, SSc, and PBC, with similar prevalence rates of each antibody across the three diseases. The specificity of ASCs in SS salivary glands suggests kinetochore complex-driven autoantibody selection, providing insight into the underlying mechanism of ACA acquisition.

CENP-A overexpression promotes aneuploidy with karyotypic heterogeneity

Roshan L. Shrestha et al.

Summary: The overexpression of CENP-A leads to chromosomal instability, affecting kinetochore integrity and microtubule attachments, increasing cell invasion, and causing aneuploidy. These results provide a molecular link between CENP-A overexpression and aneuploidy, suggesting karyotypic heterogeneity may contribute to the aggressive phenotype of CENP-A-overexpressing cancers.

Causes and consequences of micronuclei

Ksenia Krupina et al.

Summary: Micronuclei are small compartments with DNA content encapsulated by a nuclear envelope, found in cancers, during senescence, and after genotoxic stress. Compromised integrity of the micronuclear envelope disrupts DNA replication and repair, exposing micronuclear DNA to cytoplasm. Micronuclei play a central role in tumorigenesis, being a source of complex genome rearrangements and promoting a cellular immune response that may contribute to cancer metastasis.
