Modeling Human Thyroid Development by Fetal Tissue-Derived Organoid Culture

Jianqing Liang et al.

Summary: This study provides a cell atlas and organoid model for human thyroid development, shedding light on the heterogeneous population of thyrocytes and its regulatory signaling. The activation of cAMP signaling was found to enhance follicle maturation and thyroid hormone secretion. Furthermore, the study reveals the importance of chromatin accessibility for hormone production.

Enhanced Canonical Wnt Signaling During Early Zebrafish Development Perturbs the Interaction of Cardiac Mesoderm and Pharyngeal Endoderm and Causes Thyroid Specification Defects

Isabelle Vandernoot et al.

Summary: The study used small molecules and genetic zebrafish models to investigate the role of various signaling pathways in thyroid specification. It was found that overactivation of the Wnt signaling pathway during early development resulted in impaired thyroid specification and severe defects in cardiovascular development. Additionally, signaling defects between cardiac mesoderm and endodermal thyroid precursors were identified as contributing to thyroid specification impairment. Rescue experiments with transient overactivation of the BMP signaling pathway partially restored thyroid specification in models with defective cardiac development.

Capture of Mouse and Human Stem Cells with Features of Formative Pluripotency

Masaki Kinoshita et al.

Summary: The study reveals that formative stem (FS) cells, a novel type of stem cells, can directly respond to germ cell induction and differentiate into somatic tissues and germline in chimeras. Unlike embryonic stem cells (ESCs) or epiblast-derived stem cells (EpiSCs), FS cells exhibit distinct transcription factor dependencies, primarily relying on Otx2.

Adult mouse and human organoids derived from thyroid follicular cells and modeling of Graves' hyperthyroidism

Jelte van der Vaart et al.

Summary: The study establishes three-dimensional organoids from adult thyroid tissue, demonstrating their ability to mimic primary thyroid tissue and displaying similar physiological functions. These thyroid organoids also exhibit clinically relevant responses to Graves' disease patient sera, providing a new method for modeling human autoimmune diseases.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.

Single-Cell Trajectory Inference Guided Enhancement of Thyroid Maturation In Vitro Using TGF-Beta Inhibition

Mirian Romitti et al.

Summary: The thyroid gland regulates metabolism and growth through thyroid hormones secreted by TFCs. Loss of TFCs causes hypothyroidism. Using single-cell RNA-Seq, we identified cell types and discovered TGF-beta as a negative regulator of thyroid maturation. Inhibiting the TGF-beta pathway improved thyroid maturation and functionality in vitro.

Insulin-Like Growth Factor Pathway and the Thyroid

Terry J. Smith

Summary: The IGF pathway plays important regulatory roles in the development, growth, and function of human tissues, intersecting with pathways mediating the actions of various cytokines, growth factors, and hormones. IGF-I influences thyroid function and can impact the effects of growth hormone and thyroid hormones on target tissues. The pathway is also involved in both normal thyroid function and pathological conditions, such as benign enlargement and tumorigenesis, as well as conditions like acromegaly. Additionally, IGF-IR has been found overexpressed in orbital connective tissues in Graves' disease and thyroid-associated ophthalmopathy, leading to the development of therapeutic medications like teprotumumab.

Transcriptomic Signature of Human Embryonic Thyroid Reveals Transition From Differentiation to Functional Maturation

Genevieve Dom et al.

Summary: The human thyroid gland starts differentiation at 3-4 weeks of embryonic development, with functional differentiation beginning at 10-11 weeks and completed by 12-13 weeks. As the thyroid matures, hormone production increases and regulatory mechanisms are established. Through transcriptomic analysis comparing embryonic and adult thyroid tissues, this study explores the transition from differentiation to maturation of thyroid function and possible genes related to congenital hypothyroidism.
