Site- and Enantioselective Manganese-Catalyzed Benzylic C-H Azidation of Indolines

This practical method enables the azidation of tertiary benzylic C-H bonds with good functional group tolerance, leading to the efficient synthesis of diverse tertiary azide-containing indolines. The generality of the method was demonstrated by applying it to secondary azide-containing indolines. The method allows for the installation of various nitrogen-based functional groups and bioactive molecules with excellent site-, chemo-, and enantioselectivity at the C3 position of indoline frameworks.

Investigations into mechanism and origin of regioselectivity in the metallaphotoredox-catalyzed α-arylation of N-alkylbenzamides

Summary: This study investigates the mechanistic aspects of the alpha-arylation of N-alkylbenzamides catalyzed by a dual nickel/photoredox system using aryl bromides. The site-selectivity of this transformation is controlled by the generation of a HAT agent through a photocatalyst and bromide ions. The addition of tetrabutylammonium bromide was found to be crucial in increasing yields and reducing the stoichiometries of the coupling partner. NMR titration experiments and Stern-Volmer quenching studies provide evidence for the complexation and oxidation of bromide by the photocatalyst, ruling out other possible reaction pathways through mechanistic probes and kinetic isotope effect analysis.

Selectivity control over aliphatic C-H bonds is crucial and factors influencing it have been discussed. This review article provides a detailed description of HAT reagents, associated mechanistic features, and selectivity patterns observed in the functionalization of increasingly complex substrates, based on an analysis of recent literature.
