ERK inhibition in glioblastoma is associated with autophagy activation and tumorigenesis suppression

Kang Yang et al.

Summary: MAPK ERK was identified as a probable autophagy-related transcriptional response during GBM growth. Inhibition of ERK in vivo and in vitro led to cell death, demonstrating its critical role in GBM proliferation and survival. Inhibiting mTOR in combination with ERK showed enhanced antitumor effect on GBM cells, suggesting a potential therapeutic approach for GBM treatment.

Ciclopirox targets cellular bioenergetics and activates ER stress to induce apoptosis in non-small cell lung cancer cells

Junwan Lu et al.

Summary: The study reveals that ciclopirox (CPX) inhibits the migration and invasion abilities of NSCLC cells by inhibiting epithelial-mesenchymal transition, impairing cellular bioenergetics, and promoting reactive oxygen species-induced endoplasmic reticulum (ER) stress-induced apoptotic cell death. In addition, in vivo studies show that CPX can significantly inhibit NSCLC growth.

ROS generation attenuates the anti-cancer effect of CPX on cervical cancer cells by inducing autophagy and inhibiting glycophagy

Hui Fan et al.

Summary: This study found that ciclopirox olamine (CPX) inhibits the proliferation of cervical cancer cells by targeting PARK7. CPX can induce cytoprotective autophagy by downregulating PARK7 to activate PRKAA1 or by accumulating ROS independently of PARK7 to inhibit mTOR signaling. Additionally, CPX treatment increases glycogen clustering and glycophagy in cervical cancer cells.

STAT3 phosphorylation at Ser727 and Tyr705 differentially regulates the EMT-MET switch and cancer metastasis

Wei-Hsin Lin et al.

Summary: The research suggests that BM-MSC plays a crucial role in cancer cell dissemination and metastasis in lung cancer models. BM-MSC-induced signaling triggers early dissemination of cancer stem cells and promotes proliferation at secondary sites. Additionally, intercellular signaling regulates the switch between migration and proliferation.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Ciclopirox and bortezomib synergistically inhibits glioblastoma multiforme growth via simultaneously enhancing JNK/p38 MAPK and NF-κB signaling

Zhipeng Su et al.

Summary: The study demonstrated that Ciclopirox (CPX) exhibits strong anti-tumorigenic activity on glioblastoma multiforme (GBM) by inhibiting cellular migration and invasion, reducing the expression of Snail and N-Cadherin. Additionally, the combination of CPX and Bortezomib (BTZ) can promote apoptosis in GBM cells through reactive oxygen species (ROS)-mediated signaling pathways.

Delineating the Switch between Senescence and Apoptosis in Cervical Cancer Cells under Ciclopirox Treatment

Anja L. Herrmann et al.

Summary: The iron-chelating drug ciclopirox (CPX) shows potential for treating cancer, specifically cervical cancer, by inducing senescence or apoptosis in cancer cells, affecting their therapy response differently. This study explores the cellular decision-making between senescence and apoptosis in cervical cancer cells. CPX in combination with glycolysis inhibitors synergistically blocks cervical cancer proliferation by modulating OXPHOS factors and glucose availability-dependent responses, providing a rationale for future treatment strategies.

NEK9, a novel effector of IL-6/STAT3, regulates metastasis of gastric cancer by targeting ARHGEF2 phosphorylation

Guofang Lu et al.

Summary: Inflammatory stimuli in the tumor microenvironment play vital roles in cancer progression, with the mechanism of inflammation-induced cancer metastasis in gastric cancer still poorly understood. NEK9 directly regulates cell motility and RhoA activation in gastric cancer by phosphorylating ARHGEF2. The IL-6-mediated suppression of miR-520f-3p leads to an increase in NEK9, GP130, and STAT3 levels, contributing to reduced overall survival of GC patients. Targeting the IL-6-STAT3-NEK9 pathway may offer a new strategy for treating gastric cancer.
