The roles of microglia and astrocytes in phagocytosis and myelination: Insights from the cuprizone model of multiple sclerosis

Monokesh K. Sen et al.

Summary: Microglia and astrocytes play important roles in the clearance of myelin debris and remyelination in human demyelinating diseases. Recent studies have shown that up-regulation of signaling proteins in microglia facilitates effective phagocytosis of myelin debris and the release of protective mediators accelerates remyelination. However, inadequate activation or recruitment of microglia, as well as the production of toxic mediators, impairs remyelination and leads to progressive demyelination.

New oligodendrocytes exhibit more abundant and accurate myelin regeneration than those that survive demyelination

Sarah A. Neely et al.

Summary: Through research on zebrafish, it was found that surviving oligodendrocytes are less efficient in generating new myelin sheaths and frequently misdirect the myelin to neuronal cell bodies, which is similar to the pathology seen in multiple sclerosis (MS). Conversely, oligodendrocytes generated after demyelination produce abundant and correctly targeted myelin sheaths, suggesting that they may also have a better regenerative potential in MS.

Targeting the Subventricular Zone to Promote Myelin Repair in the Aging Brain

Arthur Morgan Butt et al.

Summary: The subventricular zone (SVZ) is a crucial germinal zone in the adult brain where neural stem cells (NSCs) generate olfactory interneurons and oligodendrocytes (OLs). The decline in regenerative capacity of OLs with aging contributes to cognitive decline and remyelination failure in diseases like multiple sclerosis (MS) and Alzheimer's disease (AD). This review discusses the differences between SVZ-derived and parenchymal OL progenitor cells (OPCs) and the challenges in studying their response to demyelination in vivo, highlighting their potential use in regenerative therapies for the aging brain.

Cuprizone feed formulation influences the extent of demyelinating disease pathology

Lillian M. Toomey et al.

Summary: The study showed that cuprizone pellets were more effective than cuprizone powder in inducing demyelinating disease pathology in mice, with better performance in certain indicators. However, both forms had similar effects on oligodendrocyte precursor cell dynamics.

Release of stem cells from quiescence reveals gliogenic domains in the adult mouse brain

Ana C. Delgado et al.

Summary: Adult neural stem cells in the ventricular-subventricular zone of mice can generate glial cells in addition to neurons, with certain domains remaining quiescent under normal conditions. Our study also uncovers an intraventricular oligodendrocyte progenitor derived from NSCs that is recruited upon injury.
