Formation of Breslow Intermediates from N-Heterocyclic Carbenes and Aldehydes Involves Autocatalysis by the Breslow Intermediate, and a Hemiacetal

Alina Wessels et al.

Summary: In this study, the mechanism of the reaction between N-heterocyclic carbenes and aldehydes under aprotic conditions was investigated using kinetic studies and DFT calculations. Different mechanistic regimes were proposed depending on the NHC-to-aldehyde ratio, with the H-shift occurring either in the NHC-aldehyde adduct or in the equilibrium between the zwitterion and hemiacetal forms. Furthermore, an H-bonded complex of the reaction intermediate with excess NHC was detected.

The Role of the Fused Ring in Bicyclic Triazolium Organocatalysts: Kinetic, X-ray, and DFT Insights

Jiayun Zhu et al.

Summary: The size of the fused ring in bicyclic triazolium salts has been found to have a significant impact on the rate of C(3) proton transfer reactions, with smaller rings resulting in higher reaction rates. The structural effects of altering the fused ring size were analyzed using XRD and computational data.

Synthesis of Highly Congested Tertiary Alcohols via the [3,3] Radical Deconstruction of Breslow Intermediates

Roger Machin Rivera et al.

Summary: This study reports a method for synthesizing particularly hindered tertiary alcohols via rearrangement reactions using [3,3]-sigmatropic rearrangements. Experimental studies suggest that the reaction proceeds through a close radical pair, leading to highly selective rearranged products.

Breslow Intermediates (Amino Enols) and Their Keto Tautomers: First Gas-Phase Characterization by IR Ion Spectroscopy

Mathias Paul et al.

Summary: The study reported in this paper focuses on the preparation of charge-tagged Breslow intermediates/keto tautomers derived from three different types of NHCs and aldehydes. Using ESI-MS IR ion spectroscopy, it was confirmed that in the gas phase, the Breslow intermediates derived from imidazolidin-2-ylidenes exist as diamino enols, while those derived from 1,2,4-triazolin-5-ylidenes and thiazolin-2-ylidenes exist as keto tautomers. This observation is consistent with findings in solution and in the crystalline state.

Kinetic and Structure-Activity Studies of the Triazolium Ion- Catalyzed Intramolecular Stetter Reaction

Christopher J. Collett et al.

Summary: Mechanistic studies of the triazolium ion-catalyzed intramolecular Stetter reaction revealed a first-order dependence on catalyst and zero-order dependence on aldehyde concentration. Catalysts with N-aryl substituents bearing electron-withdrawing groups showed higher reaction rates, while a concurrent, NHC-independent substrate isomerization with a first-order dependence on aldehyde concentration was also observed. The reported data suggest that deprotonation to form the Breslow intermediate is the turnover-limiting step in this process.

Fragmentation and rearrangement of Breslow intermediates: branches to both radical and ionic pathways

Ming-Hsiu Hsieh et al.

Summary: The paper discusses the key role of Breslow intermediates in N-heterocyclic carbene-catalyzed reactions, and presents different ionic and radical mechanisms depending on the protonation state. By analyzing the reactions of thiamin analogues and N-allyl benzothiazole Breslow intermediates, the study provides insights into the distinct pathways associated with specific protonation states and chemical structures of NHCs.

Tale of the Breslow intermediate, a central player in N-heterocyclic carbene organocatalysis: then and now

Monika Pareek et al.

Summary: N-Heterocyclic carbenes (NHCs) are popular organocatalysts known for their umpolung reactivity on aldehydes and ketones. Recent research has focused on gaining insights into the formation and characterization of Breslow intermediates, which play a central role in organocatalytic reactions.

Umpolung coupling of pyridine-2-carboxaldehydes and propargylic carbonates via N-heterocyclic carbene/palladium synergetic catalysis

Weiyang Bi et al.

Summary: The umpolung cross-coupling reaction of pyridine-2-carboxaldehydes and propargylic carbonates was successfully achieved for the first time through N-heterocyclic carbene/palladium cooperative catalysis. The judicious selection of the palladium catalyst, ligand, and N-heterocyclic carbene resulted in regioselective formation of propargylic ketones.

Kinetic and structure-activity studies of the triazolium ion-catalysed benzoin condensation

Richard S. Massey et al.

Summary: Steady-state kinetic and structure-activity studies were conducted on a series of six triazolium-ion pre-catalysts for the benzoin condensation, providing quantitative insight into the role of N-aryl substitution in a polar solvent environment. The observed increase in catalytic activity and rate constants with electron withdrawing N-aryl substituents supports the optimal synthetic outcomes reported under catalytic conditions. The levelling of rate constants at high aldehyde concentrations suggests a change in rate-limiting step, and a positive Hammett rho-value indicates the build-up of electron density adjacent to the triazolium N-Ar in the rate-limiting step.
