Design, Synthesis, and Biological Evaluation of (+)-Camphor- and (-)-Fenchone-Based Derivatives as Potent Orthopoxvirus Inhibitors

Summary: In this study, a library of compounds based on camphor and fenchone was synthesized and potent antiviral compounds were identified. The substituent at the para-position of the aromatic ring and the nature of the linker were found to be significant factors for antiviral activity. Compounds with para-substituted aromatic and cyclohexane rings showed the highest antiviral activity. Hydrazone and amide groups were more favorable linkers than ester groups. Compounds 3b and 7e exhibited high antiviral activity and inhibited the late processes of the orthopoxvirus replication cycle, with the p37 viral protein identified as a possible biological target.

Summary: The molecular structure and crystal packing of new aryl and alkyl N-(adamantan-1-yl)amides derivatives A1-A7 were analyzed in this study. Different types and quantities of hydrogen bonds and H...H contacts were found in the crystals of these compounds. The contribution of O...H contacts to the Hirshfeld surface is higher for alkyl adamantylamides than for aryl derivatives, while the packing index is lower for the alkyl derivatives.

Monkeypox: A Comprehensive Review

Summary: The multi-country monkeypox outbreak in humans in 2022 has added a new public health challenge on top of the ongoing COVID-19 pandemic. The disease has spread to 104 countries across six continents, with North America and Europe being the most affected. The monkeypox virus (MPXV) has been known since 1959 and has been reported to be virulent among humans since the 1970s, primarily in endemic countries in West and Central Africa. However, the disease has re-emerged at an unprecedented rate in 2022, raising concerns about its human-to-human transmission and community spread in non-endemic regions. It is crucial for healthcare workers, public health policymakers, and the general public to be well-informed about this neglected viral disease. This comprehensive overview provides information on the epidemiology, etiology, pathogenesis, clinical features, diagnosis, and management of monkeypox. It also discusses preventive and control measures, the latest vaccine developments, and future research areas for this re-emerging viral disease that has been declared a public health emergency of international concern.

Adamantane derivatives as potential inhibitors of p37 major envelope protein and poxvirus reproduction. Design, synthesis and antiviral activity

Summary: Smallpox, caused by the variola virus, has been eradicated, but other poxviruses still pose a threat to humans and animals. The p37 protein is a potential target for new drugs targeting poxvirus infections. By studying the p37 protein and using virtual screening and molecular docking, new inhibitors of orthopox reproduction have been discovered, showing promise for future antiviral treatments.
