Summary: In this study, increased TMEM40 expression was found in CSCC tissue samples compared to normal skin, and was associated with large tumor size in CSCC patients. Functional assays in vitro demonstrated that TMEM40 was involved in regulating cell growth in A431 and SCL1 cells by affecting the cell cycle and apoptosis. Silencing TMEM40 in these cells resulted in cell cycle arrest and increased apoptosis. Additionally, migration and invasion were significantly reduced after silencing TMEM40 expression in CSCC cells. These findings suggest that targeting TMEM40 could potentially inhibit CSCC development and serve as a therapeutic target for CSCC.
