Immunobiology and pathogenesis of hepatitis B virus infection

Matteo Iannacone et al.

Summary: HBV is a hepatotropic virus that can cause persistent infection and serious liver diseases. Immune responses, particularly CD8(+) T cells, play a crucial role in viral clearance and liver pathology.

Ubiquitous expression of HBsAg from integrated HBV DNA in patients with low viral load

Marie-Anne Meier et al.

Summary: Integrated HBV DNA can contribute to HBsAg production independently of viral replication, and transcribed HBV integration can extend to the entire liver in some HBe-negative patients, leading to ubiquitous HBsAg expression. This questions the clinical utility of HBsAg as a surrogate marker for viral replication in patients with low serum viral loads.

Animal Models of Hepatitis B Virus Infection-Success, Challenges, and Future Directions

Yongzhen Liu et al.

Summary: Chronic hepatitis B virus (HBV) infection affects over 250 million people globally and poses a significant risk for severe liver diseases, yet progress in research has been hindered by species tropism and lack of suitable animal models. However, development of surrogate animal models in recent years has greatly advanced HBV research in vivo.

Bioengineered Liver Cell Models of Hepatotropic Infections

Francisca Arez et al.

Summary: Hepatitis viruses and liver-stage malaria are major liver infections causing high morbidity and mortality worldwide. The development of disease models for studying infection is hindered by the limited tropism of major human hepatotropic pathogens. Bioengineered cell models exhibit better performance in simulating the human liver microenvironment compared to conventional two-dimensional cell models, and are crucial for drug development and understanding the molecular mechanisms of infection.

Hepatitis D virus in 2021: virology, immunology and new treatment approaches for a difficult-to-treat disease

Stephan Urban et al.

Summary: Approximately 5% of individuals infected with hepatitis B virus (HBV) are coinfected with hepatitis D virus (HDV), leading to unfavorable outcomes such as liver cirrhosis, liver failure, and hepatocellular carcinoma. Recent advancements in understanding HDV virology and immunology have facilitated the development of specific antiviral drugs and potentially immunotherapeutic strategies. Novel treatment options, including the entry-inhibitor bulevirtide and prenylation inhibitor lonafarnib, show promise in making HDV a less difficult-to-treat virus.

Interferon lambda 4 impairs hepatitis C viral antigen presentation and attenuates T cell responses

Qian Chen et al.

Summary: The genetic variant in the IFN-lambda 4 gene is associated with a poor prognosis of hepatitis C virus infection. The IFN-lambda 4 protein induces ER stress, inhibiting the presentation of HCV epitopes to CD8(+) T cells.
