Summary: This study presents five new cases of ectomesenchymal chondromyxoid tumors (EMCMT) of the tongue, including the first two extracranial/extrathoracic cases. The tumors showed diverse histological and immunohistochemical features, but all cases had the same RREB1::MRTFB fusion detected by targeted RNA sequencing. These findings expand our understanding of the topographic spectrum and heterogeneity of RREB1::MRTFB-rearranged neoplasms.

Summary: The VGLL3 fusion plays an important role in the tumorigenesis of a distinctive low-grade myogenic neoplasm with a predilection for the head and neck. Immunohistochemistry shows consistent expression of desmin, myogenin, and myoD1 in these tumors. Patients are usually managed surgically, with some receiving adjuvant radio- or chemotherapy. Further research is needed to fully understand the characteristics and therapeutic strategies for this tumor, which generally exhibits a benign behavior.
